Caragana opulens Kom.and Caryopteris mongholica Bungeare both important wild native plants with their own advantages in northwest of China.The purpose of this paper is to use asexual propagation technology to solve the dilemma of sexual reproduction of two kinds of plants,to fill in the blank of tissue culture of two kinds of plants,and to provide a theoretical basis for asexual propagation of other local wild drought resistant plants,to a certain extent,it can protect the diversity of local wild plants and realize the introduction of landscape greening.Based on the advantages of tissue culture,this paper took the mature seeds of Caragana opulens Kom.and the tender stems of Caryopteris mongholica Bunge as explants,preliminary established the tissue culture system of two kinds of wild native plants,filled in the relatively empty white state of tissue culture of two kinds of wild plants through the method of tissue culture,and well preserved the two kinds of wild plant germplasm resources.Combined with the statistical analysis method,the objective analysis of the data generated in the test process makes the test results more scientific,in order to provide basic data for the protection and rapid propagation of wild native plants.The main results of this study are as follows:(1)Disinfection treatment:the best way to treat the explants of Caragana opulens Kom.is 75%alcohol for 30 s and 2%sodium hypochlorite for 15 min,the survival rate is 61.09%.The best way to treat the explants of Caryopteris mongholica Bunge is 30 min of washing under water,75%alcohol for 30s and 2%sodium hypochlorite for 3 min,the survival rate is 99%.(2)In the germination pathway of Caragana opulens kom.,MS+6-BA 0.50 mg/L+NAA 0.05 mg/L was the best medium with the highest induction rate of 78.84%;MS+NAA 0.05 mg/L+6-BA 0.50 mg/L was the best medium with the best proliferation coefficient of 3.30.6-BA and NAA have significant effects on induction and proliferation,and determine the optimal level of factors.The best rooting medium was 1/2 MS+IAA 0.10 mg/L,the rooting rate was 63.16%.(3)In the way of callus induction from Caragana opulens kom.seeds,MS+6-BA 0.50 mg/L+2,4-D 0.50 mg/L was the best medium,and the highest induction rate was 75%;MS+2,4-D 0.50 mg/L+ 6-BA 0.50 mg/L was the best medium,and the multiplication coefficient was 3.00;MS+6-BA 0.50 mg/L+KT 2.00 mg/L+NAA 0.50 mg/L showed green bud on 5 days,but then there is no cluster of buds.(4)In the budding pathway of the young stem segment of Caryopteris mongholica bunge,MS+6-BA 1.00 mg/L+ NAA 0.30 mg/L+ GA 0.02 mg/L was the best medium,and the highest induction rate was 99%;MS+6-BA 1.0 mg/L+ NAA 0.3 mg/L+GA 0.02 mg/L was the best medium,and the best multiplication coefficient was about 12.00.6-BA,NAA and GA had significant effect on proliferation,and determine the optimal level of factors.MS+IBA 0.3 mg/L+GA 0.15 mg/Lwas 89.5%.The survival rate was 77%.IBA and NAA have significant effect on rooting rate,and determine the optimal level of factors.(5)Acclimatization:the survival rate of the tissue culture seedlings of Caragana opulens Kom.were 90%respectively when they were transplanted on the substrate of peat soil:Perlite:vermiculite=6:3:1.The survival rate of the tissue culture seedlings of Caryopteris mongholica Bunge was 77%when they were transplanted on the substrate of peat soil:Perlite:vermiculite=6:3:1.
