Asylnbiotic propagation and the establishment of rapid propagation system is key to cross-breeding, which have great significance in nurturing new varieties of orchids. This thesis, to study on hybrid seed germination, utilizes seeds born by Cymbidium kanran x Cymbidium ensifolium as materials. The experiment systematically studied on the relationship between the rhizomes used as materials and some factors such as basal medium, hormone, activated carbon and so on. It can help to understand how the mechanism affects the proliferation differentiation and rooting of hybrid kanran, and it has established a viable and efficient tissue culture technology of hybrid kanran. Based on that, the changes of physiology and biochemistry in development stages of differentiation and the parents which germinating hybrid kanran were preliminary studied. The results are as follows:1. System of rapid propagation of hybrid kanran:In the system of asptic propagation, orchids experienced through the seed to the protocorm and rhizomes to the plantet in differentiation stages. The optimum medium for seed germination of hybrid kanran is 1/2MS+6-BA 0.5 mg/L+NAA 1.0 mg/L+AC 1.0 g/L+peptone 2.0 g/L+agar 7.0 g/L+sucrose 30.0 g/L. The best medium of rhizomes for proliferation is MS+6-BA 1.0 mg/L+NAA 5.0 mg/L+tryptone 2.0 g/L+AC 1.0 g/L+sucrose 30.0 g/L+agar 7.0 g/L. The best differentiation medium is B5+6-BA 3.0 mg/L+NAA 0.4 mg/L+tryptone 2.0 g/L+sucrose 30.0 g/L+agar 7.0 g/L. The best rooting medium combination is B5+AC 0.5 g/L+NAA 2.0 mg/L+tryptone 2.0 g/L+sucrose 30.0 g/L+agar 7.0 g/L.2. Physiology and biochemistry of hybrid kanran: The results of the parameter of hybrid kanran in developmental stages are as follows:① In different periods of differentiation, physiological and biochemical indicators have changed dramatically.② The soluble sugar and soluble protein were increased after the first decrease and the change of chlorophyll content likes "N" type. It is also shown that both of them were closely related to the changes of chlorophyll content.③ With the advance of the development process, POD and SOD had a general downward trend, while the CAT activity had rised. they collaborated to maintain the balance of plants oxidation system.Besides, in comparison with parents, physiological indices of the hybrids were between their parents.④ Content of chlorophyll, soluble sugar and soluble protein, CAT activity:female parent> filial generation> male parent;② SOD activity, POD activity:male parent> filial generation> female parent.
