Study On The Technique System Of Tissue Culture In Cymbidium Kanran Makino

The young stem tips of Cymbidium Kanran Makino were used for explants in the study on the technique system of micropropagation.The main objects of the study were protocorms and rhizomes induction,proliferation,plantlet rooting and transplanting.The main results were follows:1.The best way of sterilizing for stem tips was as follows:immersing in 75% alcohol for 30s rinsing 2-3 times with sterilized water-disinfecting in 0.1% mercuricchloride solution for 10 min–rinsing 4-5 times with sterilized water–removal of bracteal leaves.2.The significant factors of inducing protocorms were NAA and the basic medium. The best inducement culture medium was MS+6-BA0.5 mg/L+NAA1mg/L+AC2.0g/L+sugar30.0g/L+agar5.0g/L,pH5.8.3.The significant factors of rhizomes multiplication were the basic culture medium,6-BA,NAA. The best proliferation medium was 1/2MS+6-BA0.5 mg/L+NAA1mg/L+AC2.0g/L+sugar30.0g/L+agar5.0g/L,pH5.8.4.The best optimizing medium for rhizomes was 1/2MS+6-BA2 mg/L+NAA 1mg/L+PE2mg/L+AC2.0g/L+sugar30.0g/L+agar5.0g/L,pH5.8.5. The significant factors of rooting culture was IBA .The best rooting culture medium was 1/2MS+IBA1mg/L+6-BA0.1 mg/L+NAA0.5mg/L+AC2.0g/L+sugar 30.0g/L+agar5.0g/L,pH5.8.6. Seedlings were foeged in the room for 5 days,before transplanted.The best soilless media for Cymbidium Kanran Makino was sphagna,the living rate can reach 93.33%.
