The purpose of this experiment was to study the affect of different treatment methods in H2O2 on expression of peroxisomal membrane protein-70 (PMP-70) in mouse embryos. By using the experimental method of indirect immunofluorescence, in order to detect the change of the quantity of peroxisomal membrane protein 70 in peroxisomes, and further understanding of the antioxidant mechanisms in embryo peroxisomes, we obtained the following results:1. By the method of 25 μmol/L H2O2 treatment with 1-cell in 30min, the in vitro group compared with 25 μmol/L H2O2 treatment group the quantity of peroxisomal membrane protein 70 of the 2-cell stage and 4-cell stage have significant difference, other cells stage have no significant difference.2. By the method of 25 μmol/L H2O2 treatment with mouse embryos in 30min before dyeing, the in vivo group compared with the in vitro group, the quantity of peroxisomal membrane protein 70 of other cells stage have no significant difference except the quantity of peroxisomal membrane protein 70 of 2-cell stage have significant difference. The in vitro group compared with 25 μmol/L H2O2 treatment group, at each cell stage showed no significant difference. However, the in vivo compared with 25μmol/L H2O2 treatment group, the quantity of peroxisomal membrane protein 70 of other cells stage have no significant difference except the quantity of peroxisomal membrane protein 70 of 2-cell stage and 4-cell stage have significant difference.3. By the method of H2O2 processing, ① It has been determined that 12.5μmol/L H2O2 has a great effect on the quantity of peroxisomal membrane protein 70 in mouse embryos. ② In control group,1-cell stage and blastocyst compared with 2-cell stage,4-cell stage,8-cell stage, the quantity of peroxisomal membrane protein 70 have significant differences, and morula stage without significant difference; 2-cell stage,4-cell stage,8-cell stage compared with every cells stage, the quantity of peroxisomal membrane protein 70 have significantly difference; morula stage compared with 2-cell stage,4-cell stage, the quantity of peroxisomal membrane protein 70 have significant difference, and 1-cell stage,8-cell stage and blastocyst stage had no significant difference. In 12.5 μmol/LH2O2, group,1-cell stage, the 2-cell stage,4-cell stage, 8-cell stage compared with every cells stage, the quantity of peroxisomal membrane protein 70 have significantly difference; morula stage compared with 1-cel stage 1 and 2-cell stage,4-cell stage,8-cell stage had significant difference, and blastocyst stage had no significant difference;4. It has been determined that the method of 25 μmol/L H2O2 treatment with 1-cells in 30min has a great effect on the quantity of peroxisomal membrane protein 70 in mouse embryos.
