Cyclophilin D (cyclophilin D, CypD), as a Cyclophilins,the members of the family.Studies show that CypD can regulate cell apoptosis by regulated the opening of MPTP.The objective of this study is to work out whether SpliCypD induced apoptosis of hemocytes and involved in inhibition cellular immunity of the host through regulated mitochondria pathway an innate immune response of Spodoptera litura tainted by infected Microplitis bicoloratus Bracovirus.First,we used a new RNAseq-based comparative analysis of the hemocytes transcriptomes of non-parasitized and parasitized S.litura that found the expression level of SpliCypD was significantly increased. In order to further validate SpliCypD expression in the S.litura larvae after parasitic hemocytes, we through the qRT-PCR method to detect the hemolymph of S.litura larva of parasitized 5th day SpliCypD transcription expression, the results show that the transcription SpliCypD rises significantly.By extracting the S.litura larva hemolymph of the parasitic 4 to 7 day for Western Blot test, the results showed that the protein levels of SpliCypD have obviously changed.So, we speculate that SpliCypD may involved in the S. litura larvae immune response was suppressed by MbBV. In order to further illustrate this point of view, we cloned SpliCypD gene from the S. litura hemocytes and constructed a eukaryotic expression vector PIZT/V5-SpliCypD, and transfected in Spli221 cells for overexpression. By Western Blot, immunofluorescence and mitochondria separation methods for testing, the results show that transfection carrier PIZT/V’5-SpliCypD can be an expression in the mitochondria. Finally, we detect SpliCypD and FiFo-ATPase combination as either interacting or not, wheth release of cytochrome c from mitochondria and activation Caspase 3 by in MbBV infection and overexpression SpliCypD to judgment SpliCypD may or may not have had apoptosis in the path of mitochondrion link. The results show SpliCypD interacts with FiFoATPase.but compared with the control group,cytochrome c from mitochondria and Cl-Caspase 3 was not significantly.For its mechanism.it should be further studied in a clinic.In conclusion, protein levels of SpliCypD were have up-regulated in S.litura hemocytes by M. bicoloratus and MbBV infection Spli221 cell; SpliCypD are in mitochondria and are interacted with FiFoATPase,and yet, the host hemocytes apoptosis induce by whether ATPase activity inhibited lead to reduced ATP production that should be further studied in a clinic.
