| Based on the former transcriptomes research data for the fetal caprine skin of 45days,55days and 65days,selected the follicle development relative gene FoxNl and FoxI3 which belong to Fox gene family as the candidate genes,in addition to select FoxEl as another candidate gene according to relative references.Analyze the variation of this 3 candidate genes mRNA and protein in the skin by the RT-PCR and Western blot techno logy, intended to make some theory foundation and provide evidence of study the Fox gene family function mechanism in regulate the follicle develop and growth.The research results as following:(1)Analyze the differentially expressed genes in fetal 45days skin,55days skin and 65days skin by bio informatics methods,found the 204 DEGs from 45days to 65days,including FoxN1 and FoxI3,in addition to select FoxEl as another candidate gene according to relative references. There fore,FoxNl,FoxI3 and FoxEl as the candidate genes to study function mechanism in regulate the follicle develop and growth.(2)The RT-PCR result shows that FoxNl and FoxEl continued to increase during the fetal stage,and on the skin of 135days of fetal this two gene expression get the highest point;FoxI3 expression increased begin 45days and in the 75days the expression get the highest point,and from the 85days begin to decrease.In mature goat skin,the FoxEl and FoxNl expression trend consistent with follicle growth cycle,the FoxI3 expressed in July and April are higher than another month,and expressed highest in July (P<0.05).(3)The western blot result shows that the protein express of FoxNl increased from 75days to 135days in fetal and get the highest in 135days;the protein express of FoxEl get the highest point in 95days and the protein express decrease from 105days.And in mature goat skin, the protein of FoxNl and FoxEl are expressed both in anagen,catagen and telogen stage of follicle growth cycle,and this two proteins expression get the highest point in September which belong to anagen. |
