This study is based on the transcriptome data from skin of Inner Mongolia cashmere obtained from previous studies,by using GO and KEGG signaling pathway analysis to find the deferentially expressed genes in implanted group and control group from February,October and December.Finally,we selected five differentially expressed genes,Wnt10b,?-catenin,SFRP1,FGF21 and TCHHL1,as candidate genes,then used q-PCR and Western blot to validated the expressions on mRNA and protein level.All these findings can provide theoretical evidence for further investigation of promoting the cashmere growth by melatonin.The main findings of this study were as following:1.Uisng the transcriptome data of February,October and December in Inner Mongolia cashmere goat which were divided into two groups:implanted group and control group to find the diffferentially expressed genes in each month by bioinformatics method.A total of 1,478 genes were found in three months which 1014 genes were up-regulated,and 464 genes were down-regulated.2.The results of q-PCR indicated that after melatonin embedding,the expression of WntlOb gene in January and March was significantly higher than control group(P<0.05),however,the expression level in February was significantly lower than control group(P<0.05).The expression of ?-catenin gene in November and January was significantly higher than control group(P<0.05),but significantly lower than control group in July(P<0.05).The expression of FGF21 was significantly higher than control group in November,January and February(P<0.05).The SFRP1gene expressed higher than control group in June and July(P<0.05),but there was no significant difference in other months(P>0.05).The expression level of TCHHL1 gene in April,June and August was significantly lower than that in control group,however,expressed higher than control group in January and February(P<0.05).3.The results of Western blot showed that after melatonin embedded,the expression level of Wnt10b protein was up-regulated during December and February,but there was no significant difference in September(P>0.05).The expression level of ?-catenin protein was significantly lower in the test group than control group(P<0.05),however,there was no significant difference in December and February between test and control groups(P>0.05).In conclusion,it can be inferred that 5 deferentially expressed genes like Wnt10b,?-catenin,SFRP1,FGF21 and TCHHL1 play important roles in promoting the development of follicle in cashmere goat which implanted melatonin.
