The Saperda populnea is kind of herbivorous coleopterous insect which distributes over north areas of our nation. These pests damage poplars over their larva period and destroy poplar collateral parts of 2 to 3 years. What’s more, it makes insect galls appear which influence the transportation of water and nutrition, and may cause drying up or breaking off of limbs. In recent years, the harm of cathay poplar longicom increases year by year, which poses a threat to poplar shelterbelts of northern areas in our country.This paper chooses populus xiaohei and populus albaberolinensis as research subject. After being damaged by cathay poplar longicorn, populus phloem will appear some defense reaction. Through dyeing treatments and metabolomic analysis, this paper explore the defense reaction after being harmed that can help us find scientific basis of the prevention and treatment of cathay poplar longicom.Experiments show that:1. Over the long period evolution with insects, plants form their own defense systems. When being damaged by pathogenic microorganisms, plant hoppers and mechanical damage, plants will appear defense reactions triggered by the exchange of potassium ion, release of calcium ion, ROS, H2O2 and secondary metabolites qualitative accumulation. Salicylic acid makes ROS and the dead cells accumulate. ROS are some O2- and H2O2. This paper use NBT to detect O2-, DAB to detect H2O2, trypan blue to detect dead cells. Dying discovery:after being affected, the enzyme’s activity of aspens will be lowered and it’s color will be darker, which means some cells die for the ultra oxygen anion and hydrogen peroxide solution. This phenomena proves that aspens will appear response after being poisoned by saperda populnea or mechanical damage.2. Through the change of populus phloem metabolome after damaged by Saperda populnea, this paper uses the method of qualitative analysis and quantitative analysis to analyze the metabolites, uses GC-MS to detect phloem metabolome of populus phloem,analyze the spectrogram and track some plant cells or metabolite. Combine the method of multivariable statistical analysis, relative ion fragmentation and NIST to retrieve, check and recognize the physiological and pathological relationship of plants. Finally, the passage get the source’s change information produced by metabolite.Then use the method of PLS to analyze and compare metabolomes of different experimental groups and matched groups. We have found the varieties of metabolic substances are mostly same but the average content of different metabolic substances have apparent difference. This experiment successfully filtered some specific metabolic substances whose average content are higher than others, found the positive and negative correlation of different metabolic substances, finally get the way that some specific metabolic substances changes the metabolic pathways. So we can know the response of aspens being poisoned.
