Preliminary Study On Saperda Populnea Virus

This article expatiated about the researching on Saperda populnea Virus. After collecting and anatomizing the galls of Saperda populnea, some black dead larvas were found out, which were not so hard-shelled. By using the method of different speed centrifugation got some extracted virus substance. And by using the method of gradating centrifugation got the purer virus. After doing infection experiment and checking under the photic microscope and electron microscope. The virus substance was figured to be a kind of polyhedrosis virus. This kind of virus take place in northern Shanxi province, infects the larvas of Saperda populnea and makes them dead. The techniques about preserving and reproducing of Saperda populnea Virus were studied. Saperda populnea Virus can be reproduced by using the way hereinafter: The remainder of insect warts after anatomizing can be used as substrate, and metal boxes can be used as cultivating boxes. Put the substrate into the cultivating boxes and spray some Saperda populnea Virus solution, and put the Saperda populnea larvae in the substrate. The Saperda populnea Virus can be reproduced in the bodies of larvae under 15~20℃. The Saperda populnea Virus should be preserved under about 4℃. The virus can also be directly preserved the insect warts in a cellar (about 7℃). The Saperda populnea Virus can keep long time alive under such condition.
