Cow Ketosis is a common metabolic disease caused by a mass of carbohydrate and fat metabolism disorders caused,the disease mainly in ketosis, ketones urine disease, breast disease is characterized by a ketone,usually hypoglycemia, elevated blood free fatty acids, fatty liver, glycogen levels decline, mainly in high-yielding dairy cows. Although ketosis can be cured, Cows also rarely cause death, but ketosis can cause decreased milk production, milk quality and reduced fecundity, secondary reproductive system diseases and endocrine disorders and other diseases, increased treatment costs, cause greater damage to the dairy industry. Timely and efficient manner is important to improve the level of development of prevention and treatment of cows milk cow ketosis.Object: In this study, Shihezi Region 96 scale dairy cows milk and serum as material, determination of milk and ketone bodies, β- hydroxybutyric acid concentration in serum, to study methods for detecting cows ketosis large-scale farms and Ketosis cows milk and serum β- hydroxybutyric acid(BHBA)concentration correlation.Methods: 1、Ketone powder method, Reagent method, ROSS method, Rothera method, Four methods qualitative detection of milk; Modified salicylaldehyde quantitative detection of cow serum, were qualitatively detected in milk; Improved quantitative assay of cow serum salicylaldehyde, defined serum ketone body content of more than 100 mg / L of ketosis cows.2、Applications phosphotungstic acid precipitation method for ketosis cows emulsion deproteinizing,then determination of milk β- hydroxybutyric acid concentration by enzyme assay,determination of ketosis cows β- hydroxybutyric acid concentration in serum by enzyme-linked immunosorbent assay, were analyzed using SPSS 10.0, the correlation coefficient pearson.Results: 1、The number of modified salicylaldehyde measured by 96 cows suffering from ketosis to40, Qualitative detection of four kinds of milk powder method accurate method ketone highest coincidence rate 85.42%.2、Ketosis cows emulsion BHBA average concentration of 1.21 ± 0.25mmol/L, Serum BHBA average concentration of 2.19 ± 0.35mmol/L, with increasing parity, milk in BHBA concentration gradually increased, the serum concentration does not significantly change BHBA; The higher milk yield, milk and serum BHBA content is gradually increased, and higher correlation coefficient between the two.Conclusion: 1 、 Ketone powder method assay ketosis cows high detection efficiency and easy operation, the false positive rate, ketone powder method for detection of ketosis as a large farms.2、In ketosis cows milk and serum β- hydroxybutyric acid correlation of 0.8102-0.9105, Thus milk β-hydroxybutyric acid can be determined whether ketosis cows by milk β- hydroxybutyric acid detection, as early diagnosis is based on cows ketosis.
