| Verticillium wilt is caused by Verticillium dahliae, the most destructive in the soil-borne in cotton process, and causes serious influence to the quality and production of cotton. The long-term use of chemical pesticides to control it will damage the ecological environment and make pathogens produce drug-resistant, and also limit the cotton industry’s sustainable development. Because people hadn’t find a effective prevention and control technology of biocontrol method, it was known as "cotton cancer". For many years research showed that fostering new varieties of resistance is the most direct and efficient strategy, and the screening and identification of the anti-source is the most important step of cultivating a new variety.The research has used Verticillium dahliae10(Vd10) to do resistance level measurement for transgenic Arabidopsis thaliana, respectively using inoculate in petri-dish and the root-dipping inoculation in soil. And have done PCR and Southern blot identification to 14 transgenic materials, the main result as follow:1) The inoculation identification results indicated that 3 materials with higher resistance, 8 materials with medium resistance, and 2 materials with no specific resistance to Vd10 equal to WT.2) Using the methods of petri-dish inoculation and root-dipping inoculation identification results showed: In petri-dish inoculation cause plant disease uniformitily, seriously and quickly, and we can clearly observed the pathogen’s process of breeding, expanding and being taken bad in plant root. Moreover, we could obtain the identification result after sowing 42 days, but the root-dipping inoculation method needed 60 days. At the same time, the study also shows that the in petri-dish inoculation allowed us to analysis nearly 50 young plants in a relatively small and complete control petri dish environment, and in greenhouse could also identify more than 10 Arabidopsis thaliana strains resistant ability identification, hence this method is a effective identification method of against pathogen. 3) The results of PCR identification indicated that 13 transgenic plants are masculine strains; and southern blot identification result showed transgenic plants have one copy specific gene of transformation at least.The result of this research will provide new anti-source for new cotton breeding work of resistance to verticillium wilt. |
