The study eatablished the in vitro plantlet regeneration of 9501 Paulownia with its leaves and petiole for test material.In addition,this study induced polyploidy with its leaf callus tissue for explant by colchicine,which will be valuable materials and reliable theory basis for deeply research and breeding sterile plant of Platanus orientalis L..The main results are as follows:(1)In vitro plantlet regeneration system for 9501 Paulownia is established.Among petioles and leaves,leaves were the best explants for vitro plantlet regeneration. The optimal media for callus、shoot and root induction for 9501 Paulownia is MS + 0.7 mg·L-1 NAA + 8 mg·L-1 BA, MS + 0.7 mg·L-1 NAA + 8 mg·L-1 BA and 1/2 MS.(2)Use double-cultured method induced tetraploid plant of 9501 Paulownia,The concentration of colchicine and treatment time produced highly significant variance to the experiment, pre-culture time produced significant variance to the experiment.The test that use double-cultured method induced tetraploid plant of Platanus orientalis L..In two factors of induction tetraploid,the concentration of colchicine produced extremely significant variance to the experiment,the treatment time produced Significant variance to the experiment.(3)The appropriate combinations for tetraploid of 9501 Paulownia induction was combination (colchicine 10 mg·L-1+treatment time 72 h+pre-culture time 8 d),and the induction rate is 18.57 percent.The appropriate combinations for tetraploid of Platanus orientalis L. induction was combination (colchicine 1000 mg·L-1+treatment time 72 h),and the induction rate is 20.9 percent.(4)Chromosome number of the tetraploid of 9501 Paulownia was 80 and that of diploid plant was 40.Results of the flow cyometric analysis showed the tetraploid plant present twice higher DNA content than the diploid plant.Chromosome number of the tetraploid of Platanus orientalis L. was 84 and that of diploid plant was 40.Results of the flow cyometric analysis showed the tetraploid plant present twice higher DNA content than the diploid plant.(5)Compared with the diploid of 9501 Paulownia,leaves of tetraploid plants were larger thicker and greener.The stomata size of tetraploid plants were significantly larger,while the frequency of stomata were reduced significantly. The tetraploid of Platanus orientalis L.is the same as 9501 Paulownia.
