| Objective:To explore the effect of NaoDeJian Decoction on Neurological Assessment and Neuroregeneration in rats after focal cerebral ischemia.Methods:The model of focal cerebral ischemia in rats was reproduced by middle cerebral artery occlusion. Rats were randomly divided into following groups including Normal group, sham group, model group, BYHWD group,high groupsã€middle dose group and low-dose group of the NDJD. All the animals received injection of bromod—eoxyuridine(BrdU)since the experiment started. which received different treatments. and then the rats were kilied at 3,7,14 day after operation. The expressions of Brdu and Brdu+/NSE+ and Brdu+/NF200+were measured by immunohistochemical staining,at each time point, and their neurological functions were observed; immunohistochemical methods were applied to detect the number of their newborn neurons.Result:1. The rats After focal cerebral ischemia model with nerve dysfunction, NDJD significantly decreased neurological functional deficits after focal cerebral ischemia. There were Not significant differences between the NDJD and BYHWD groups.2. There is a certain BrdU positive cells in normal brain, after cerebral ischemia BrdU positive cells were increased,and NDJD decoction can significantly increase the expression of BrdU. There were significant differences between the Drug group and model group at 3,7 and 14 d (P<0.05). High groupã€middle dose group of the NDJD Compare to the BYHWD group were not significant differences (P>0.05)3. After cerebral ischemic. Brdu+/NSE+ double-staining cells were increased at 3,7d and the basic Same with the model group at 14d. NDJD decoction and BYHWD can significantly increase the expression of Brdu+ /NSE+ double-staining cells. There were significant differences between the NDJD groups and model group at 3,7 and (P<0.05). High groupã€middle dose group of the NDJD Compare to the BYHWD group were not significant differences (P>0.05)4. After ischemic, there was few Brdu+/NF200+ double-staining cells was in subcortex, hippocampul on 3d,number of double-staining cell reached peak on 7d. NDJD decoction and BYHWD can significantly increase the expression of Brdu+/NSE+ double-staining cells. There were significant differences between the NDJD groups and model group at 3,7 and 14 d (P<0.05). High groupã€middle dose group of the NDJD Compare to the BYHWD group were not significant differences (P>0.05) 5. There is linear between functional index and number of new neuron. MathType or Equation Editor is y=0.866X+0.90.Conclusion:1.NDJD significantly decreased neurological functional deficits after focal cerebral ischemia. There is similar effects between the NDJD and BYHWD.2. NSCs were activated, proliferated and differentiated into neuron after cerebral ischemic, There is Positive relation between neurogenesis and neural functional index. NDJD can expand the proliferation and neurogenesis, There were Not significant differences between the NDJD and BYHWD groups. |
