Effect Of Naodejian Decoction On IL-6,IL-1β And TNF-α Expression And Cell Proliferation In Rats After Focal Cerebral Ischemia

Objective:To explore the effect of Naodejian Decoction on inflammatory factors and cell proliferation in rats after focal cerebral ischemia.Methods:Rats were randomly divided into following group including shame control,ischemic, ischemic with BYHWD,ischemic with ND High-dose group, ischemic with ND Medium-dose Group, ischemic with ND Low-dose group, focal cerebral ischemic model was reproduced by middle cerebral artery occlusion, Animals were received BrdU-labeled and killed at 1,3 and 7 day point after operation.5 rats were randomly taken from each group and calculated infarct size ratio by TTC,5 rats were calculat the ratio of wet and dry brain. Inflammatory factors were measured by ELISA,RT-PCR and Brdu were measured by immunohistochemical staining.Result:1.ND could decrease infarct volume and edema after focal cerebral ischemia. 2.ND could reduce the expression of IL-1β, and TNF-αin protein and mRNA levels after focal cerebral ischemia.3.ND could control the expression of IL-6:Could reduce its secretion obviously in the beginning and 7d,and promote its secretion obviously in 3d.4.ND could promote the expression of BrdU positive cells.Conclusion:1.ND has the same efficacy as BYHWD on decreasing infarct volume and edema。2.ND has the same efficacy as BYHWD on control inflammatory factors expression in protein and mRNA levels.The result show that it protect brain by control inflammatory factors expression.3.ND has the same efficacy as BYHWD on tissue self-repair by promote cell proliferation after focal cerebral ischemia.
