Objective:The purpose of this study is to explore the mechanism of Sijunzi Decoction on neuroprotective effects of focal cerebral ischemia-reperfusion injury.Methods: 48 SD male rats were randomly divided into control group,model group,nimodipine group and Sijunzi decoction group.The rat FCIR model was prepared by middle cerebral artery occlusion(MCA).The control group was not modeled.The rats in the control group and the model group were treated with normal saline for 14 days.Nimodipine and Sijunzi decoction were treated with Sijunzi Decoction for 14 days.Evaluation of Neurological Function in FCIR Rats by Zea-Longa Grade 5 Scoring Method.The rats in each group were sacrificed after 14 days of intragastric administration.After the brain was taken,the rats were treated with paraformaldehyde and then fixed.The histopathological changes of the sections were observed by HE staining,the expression of laminin,collagen IV,tissue inhibitor of metalloproteinase-1and matrix metalloproteinase-9 were detected by immunohistochemistry.Result:1.Different groups of rats limb nerve function score:Compared with the control group,the neurological function scores of the model group were significantly lower than those of the control group(P <0.01),and the neurological function scores of the model group were significantly lower than those of the control group There was no significant difference between the two groups(P> 0.05).2.Effects of Sijunzi Decoction on Expression of LN and Col IV Proteins in Cerebral Cortex of FCIR Rats The positive expression of LN in cerebral cortex of model group was significantly lower than that of control group(P <0.01).The positive expression of LN in Sijunzi Decoction and Nimodipine group was significantly higher than that in model group(P <0.01).The positive expression of COL IV in the cortex of the model group was significantly lower than that in the control group(P <0.01).The positive expression of COL IV in Sijunzi Decoction and Nimodipine group was higher than that in the model group(P <0.01)3.Effects of Sijunzi Decoction on Expression of TIMP1 and MMP9 in Cerebral Cortex of FCIR Rats The expression of TIMP1 and MMP9 in the model group was higher than that in the control group.The positive expression of TIMP1 and MMP9 in the Sijunzi Decoction group and the nimodipine group was decreased.The positive expression of TIMP1 and MMP9 in cerebral cortex of model group was significantly higher than that of control group(P <0.01).The positive expression of TIMP1 in Sijunzi Decoction and Nimodipine group was significantly higher than that in model group(P <0.01),while MMP9 positive expression was significantly lower than that in model group(P <0.01).4.Effects of Sijunzi Decoction on HE staining of cerebral cortex in FCIR rats There were no obvious abnormalities in the neuronal cell morphology of the control group.The neuronal cells in the model group showed typical ischemic injury.The neuronal cells in the Sijunzi Decoction group and the nimodipine group also showed changes in the ischemic injury,Group slightly,arranged more neat than the model group,part of the nuclear irregular.Conclusion:1.According to the Zea-Longa grade 5 score,the neurological score of rats in each group was judged to be successful.The rat FCIR model was successfully prepared by middle cerebral artery occlusion(MCA)2.The use of uniform diameter of the fish line,as well as from the external carotid artery inserted into the bolt can improve the success rate and survival rate;3.Sijunzi Decoction can reduce the pathological changes of cerebral cortex of FCIR rats,play a neuroprotective effect;4.Sijunzi decoction may play a neuroprotective role by promoting the expression of LN and Col IV in FCIR rats;The expression of TIMP1 in the cerebral cortex of FCIR rats may inhibit the expression of MMP9 and play a neuroprotective effect.In summary,Sijunzi decoction on FCIR rat cerebral cortex has a certain degree of neuroprotective effect,and Ru Yang day after the close relationship,and its further mechanism to be confirmed.
