| Objective:By observing the influence of the Qu Shi Chu Bi method on rat modelof acute gout arthritis of jiont symptoms， serum IL-1and IL-1R，and theexpression of inflammatory cytokines(IL-1β、 IL-1R),and the signalingpathway protein MyD88，and the unit area of lymphocytes and macrophages toexplore the effect of treatment of acute gout arthritis of the mechanisms.Methods:To randomly divided40SPF Wistar rats into blank group, model grou-p, colchicine group, traditional Chinese medicine Low-dose group groupand high-dose group,8rats in each group. Injection uric acid sodium cry-stals in rat ankle to establish acute gouty arthritis model besides theblank group. Injection the same volume of saline in rat ankle of the bl-ank group. Fed the rats with medicines after modeling2hours. The blan-k group and model group be fed with distilled water1ml/100g. Observat-ion of the inflammation index and dysfunction index of rats after mod-eling8hours,72hours. Compare the1day before the modeling, two hou-rs after modeling and after modeling the first three days, seven daysof left rear foot volume. Medicated the rats after3hours and detectio-n in serum levels of IL-1and IL-1R in the first7days after modeling.Drawn right ankle joint of the rats and Observation the expression of ra-t articular inflammation cytokine IL-1β, IL-1R,and the signaling pathw-ay protein MyD88and the number of inflammatory cells in unit area ofsoft tissue in the rats joint.Results: Comparison of inflammation and joint dysfunction in rats expone-ntial integral, model group, the most points，and compare with the treat-ed groups has significant difference (p＜0.05). Compare the8hours and72hours after the modeling, points in each group is lower, and has si -gnificant difference (p＜0.05). Compare the degree of joint swelling,mo-del group is great higher than all treated groups, and has great signif-icant difference (p＜0.01). Compare colchicine group and low-dose groupwith high-dose group, and has significant difference (p＜0.05). Betwee-n the colchicine group and low-dose group has no significant difference(P＞0.05). Compare the levels of IL-1and IL-1R in serum, the model grou-p was significantly increased compare with the other4groups（P＜0.05）and has no significant difference beween all treated groups(P＞0.05). Co-mpare the IL-1β,IL-1R,MyD88in the value of the average optical densit-y，the four groups were higher than the blank group and has great signif-icant difference（P＜0.01）. Model group is greater than all treated gr-oups and has significant difference (p＜0.05). Compare colchicine groupand low-dose group with high-dose group, and has significant difference(p＜0.05). Between the colchicine group and low-dose group has no sign-ificant difference(P＞0.05).Compare the per unit area of lymphocytes,macrophages of groups, the four groups were higher than the blank groupand has great significant difference（P＜0.01). Model group is greaterthan all treated groups and has significant difference (p＜0.05). Compar-e colchicine group and low-dose group with high-dose group of per unitarea of lymphocytes, and has significant difference (p＜0.05). Betweenthe colchicine group and low-dose group has no significant difference(P＞0.05).Conclusion:1.The Qu Shi Chu Bi method can improve the arthritis symptoms of the ratmodel of acute gouty arthritis.2. The Qu Shi Chu Bi method can though inhibit the signaling pathwa-y protein MyD88in joint organization to blocking Toll/IL-1R-MyD88sign-aling pathway,and inhibit the activation IL-1R and expression of IL-1β,and reduce the number of joints within lymphocytes and macrophages. So as to achieve the the role of regulation the immune of acute antii-nflammatory and protect joints.3.The Qu Shi Chu Bi method can effective in treatment of acute goutyarthritis and it could be the basic treatment principles. |
