Objective To investigat the expression of phosphorylated Janus kinase2-JAK2),phosphorylated signal transducer and activator of transcription3(P-STAT3) andcaspase-3after acute spinal cord injury(ASCI) in rat.Method A total of90adult male SD rats, weighing225-275g, were selected randomlyand divided into the sham operation(n=30),SCI(n=30) and AG-490+SCI(n=30) groups; thenput them into the subgroups randomly, which are4h,12h,24h,72h and168h(n=6). Acutespinal cord injury rat’s model were established by modified Allen’s weight drop method. Thesham operation group just underwent T9laminectomy without injuring spinal cord. The ratsshould be sacrificed at corresponding time after operation and spinal cord tissues should betaken from them. Hematoxylin and eosin(HE) staining observe tissues morphologicalstructure and immunohistochemistry method detect the expressions of P-JAK2,P-STAT3andcaspase-3.Result The SCI model was set up and the samples of spinal cord tissues were acquiredsuccessfully. HE staining showe that the stages of serious spinal cord tissues damage were72h after ASCI, rather than4h. The low expressions of P-JAK2,P-STAT3and caspase-3areobserved in the sham operation group by immunohistochemistry method. In the SCI group,the expressions of P-JAK2,P-STAT3and caspase-3is obviously higher than the shamoperation group at all time points(p<0.01). P-JAK2and P-STAT3significantly increase from4h, peaking at12h after injury, then gradually decrease.Caspas-3rise rapidly, peaking at72h after ASCI. The expression of P-JAK2,P-STAT3and caspase-3in AG-490+SCI group werelower than in SCI group(p<0.01).Conclusion These findings suggest that JAK2/STAT3signaling pathway is abnormallyactivated in ASCI, and this pathway may be play an important role in apoptosis after SCI.
