| Objective To observe the effect of berberine on the recovery of neural function in mice after spinal cord injury.And the mechanism of berberine on anti-inflammatory through JAK2/STAT3 pathway was also be explored.Methods The animal model of spinal cord injury was prepared by Allen,s method.Sixty mice were randomly divided into three groups,20 mice in each group:Sham group(sham operation),SCI group(spinal cord injury)and Ber group(berberine treatment).The mice in Ber group received daily tail vein injection of berberine(5mg/kg)for 7 consecutive days.The BMS scoring method was used to detect the motor function of the hind limbs of mice in each group.The walking,trunk movement and coordination of the mice were observed before and 1,3,7,14,21 and 28 days after operation,and the scores were recorded.HE staining was used to observe the pathological morphological characteristics of spinal cord tissue.The expressions of Neu N and GFAP were observated by immunohistochemical staining,and the levels of Neu N and GFAP were detected by measuring the number of positive cells or optical density.The protein content of IL-6,TNF-α,IL-1β and IL-10 in spinal cord samples were detected by enzyme-linked immunosorbent assay(ELISA).Real-time quantitative PCR(q RT-PCR)was used to detected the expression of TNF-α m RNA,IL-6 m RNA,Arg-1 m RNA and TGF-β1 m RNA in spinal cord samples of mice in each group.The protein levels of P-JAK2,JAK2,P-STAT3,STAT3 were detected by Western blot.Results1.The behavioral scoring results showed that berberine significantly improved the motor function of mice after spinal cord injury.From 7 to 28 days after injury,there was a significant difference between the Ber group and the SCI group.2.The results of HE staining,Neu N and GFAP immunohistochemical staining showed that berberine improved the pathological morphology of the injured spinal cord,increased the survival of neurons,and inhibited the activation of astrocytes at 28 days after spinal cord injury.3.Three days after spinal cord injury,ELISA results confirmed that berberine reduced the secretion of pro-inflammatory factor include TNF-α、IL-1βand IL-6,increased the secretion of IL-10,which is the anti-inflammatory factor.The results of q RT PCR experimental confirmed that berberine reduce the expressions of TNF-α m RNA and IL-6 m RNA,but increase the expressions of Arg-1 m RNA and TGF-β1 m RNA.These results indicated that berberine inhibit inflammatory response during the acute phase of spinal cord injury.4.The results of Western blot experiments confirmed that berberine inhibit the activation of JAK2/STAT3 signaling pathway during the acute phase of spinal cord injury.Berberine regulated inflammatory response through JAK2/STAT3 signaling pathway,and promoted the recovery of motor function of SCI mice.ConclusionsBerberine reduces inflammatory response,increases neuronal survival,improves pathological morphology of spinal cord tissue,and promotes motor function recovery in mice after spinal cord injury through the JAK2/STAT3 signal pathway. |
