| Objective To investigate the SNPs of INH and FSHR geneã€the level of reproductivehormones and autoantibodies of Ningxia chinese women with premature ovarian failure(POF);to analyze the relationship between the SNPs of both and POF, providing the theoryevidence for the treatment of POF women, in order to impove the quality of their life.Methods63patients with POF and58controls at the Center for Reproductive Medicine inGeneral Hosptial of Ningxia Medical University.To analyze chromosome karyotype in POF;to detect reproductive hormones by chemiluminescence immunoassay(CLEIA) andautoantibodies by enzyme-linked immunosorbent assay (ELISA). INHA was amplified byPCR-RFLP, INHβBã€Ala307Thr and Ser680Asn of FSHR detected by direct sequencing. t-test,chi-square test were used for statistical analyses by performing SPSS17.0software.Results (1)The age of menarche of POF group was significant earlier than that in controls(13.87±1.69vs.14.72±2.53, P<0.05).(2)The values of FSHã€LH were also significant higherthan controls(73.85±24.41vs.6.14±2.02,50.80±17.83vs.4.05±1.66, P<0.01). The measuresof PRL〠E2〠T were significant lower in POF group than controls (8.39±4.39vs.13.08±5.45,11.22±8.33vs.42.59±14.18,2.53±16.08vs.42.85±13.52, P<0.01).(3) Positiverate of AOAbã€ACA and TGAb were significant higher in POF group than in controls(19.05%vs.1.72%,14.29%vs.3.45%,11.11%vs.0%, P<0.05).(4)The rate of abnormalkaryotype was4.76%which were46,XX,16qh+ã€45,Xi and46,Xi(xp),14qh+, respectively.(5)The mutation rate of POF group of Ala257Thr and Por365Por were higher than controls(12.69%vs.4.76%,30.16%vs.22.41%, P>0.05).(6)There was significant difference of the mutation rate and allelic frequencies with the exon10of FSHR Ala307Thr and Ser680Asnpromoter variants between Hui POF patients and Hui controls(81.6%vs.47.9%,86.8%vs.64.6%, P<0.05).(8)The interaction between INHA and FSHR Ala307Thr and Ser680Asnpromoter variants were not exist.Conclusion (1) The age of menarche is earlier in POF group than controls and the levels ofFSHã€LH are elevated in patients and low levels in PRLã€E2ã€T, suggest that H-P-O aix existsdefference scince adolescence.(2) Positive rate of AOAbã€ACA and TGAb elevated in POF,demonstrate that the women with autoimmune disorders and abnormal chromosome got thehigh risk to suffer from POF.(3)The mutation rate of patients of Ala257Thr are higher thancontrols,revealed that the mutation is involved in the pathogenesis of early premature ovariandescend.(4)The Ala307Thr and Ser680Asn of FSHR are no difference between POF andcontrols.However, the A allelic frequencies of Ala307Thr and Ser680Asn is higher than Gallelic frequencies, mention the variants are associated with Hui patients with POF.(5) Theinteraction between INH and FSHR are not exist, it is possible impact from the differentaspects of POF. |
