Objective: The premature ovarian failure(POF),as one of the stubborn disease in Gynecological diseases,is a kind of the decay of the ovarial function before the woman is 40.It shows to last amenorrhea, the estrin descends and the antophysin heightens, accompanying with the symptoms of climacteric syndrome.At present,we have not known its etiological factor,and the treatment is not satisfied.The hormonal therapy is usually used in western medicine,but it can bring to a series of side effect, and it does not get at the root. We based on traditional medicine, have presumed that the etiological factor and pathogenesis of this kind of disease be renal deficiency and hemophthisis.So we base on supporting blood and invigorating kidney and makes out a prescription named as"Yangxuebushenfang".By the clinical usage of this prescription,the satisfied effect is shown.This study is to approach the pathogenesis further that Yangxuebushenfang treats POF by observing oophoric histomorphology and detecting blood biochemical indicator at two aspects of clinical observation and experiment study.MethodsPart 1 The Emperiment Study of YXBSF Treating Premature Ovarian FailureIn experiment study, 50 healthy, female SD rats were classified into two groups randomly. One is control group, the other is model group which has 35 rats .the estrous cycle was regularity to set up the model of POF through the autogenous immunization.When the model set up successfully,killed 5 rats in each group. the 30 rats in model group would be divided into 3 groups randomly the model control group, the traditional Chinese medicine group and the western medicine group, and each group included 10 rats. The rats in the traditional Chinese medicine group were fed with Yangxuebushenfang by intragastric administration, the rats in the western medicine group were intragastric administration with the man-made cycle method, the rats in the model group and control group were fed with normal food. At both before and after making model and both before and after treating, we observe the general state of health, the level of gonadal hormone in serum and the appearance changes of the ovaries cytology of the rats in each group.Part 2 The Clinical Study of YXBSF Treating Premature Ovarian FailureIn clinical research 40 women with POF are chosen .We classified the 40 patients of POF into 2 groups, 20 women with POF are chosen as treated group; 20 women with POF are chosen as control group. The treated group is treated with Yang Xue Bu Shen Fang . The control group is treated with west medicine. Symptoms are observed pre and post treatment after treating them 3 months; Electrochemistry radiation method has been used to examine the levels of the follicle stimulating hormone(FSH), the luteinizing hormone(LH) and the aquadiol (E2)in the patients serum both before and after treatment.ResultPart 1 The Emperiment Study of YXBSF Treating Premature Ovarian FailureIn experiment study, pre-modeled rats activity is multifarious , repond intelligent ,hair color sheen.feel attracted a period 4-5day once .rats activity degree is little after building mold,repond cool,hair have no sheen. Those feeling attracted the period proiongs even disorder. LH level in the mold group is not decreased notably after modeled compared with before(p>0.05), where FSH level increase significantly compared with pre-modeled(P<0.05).E2 level decline significantly compared with pre-molded(P<0.05).Though the microscopy opticaling , growing follicecs and corpus luteums reduced significantly in POF model rats.Where atretic follicles increase significantly. After Chinese medicine treated, rats activities rise multifarious, hair color sheen. FSH level in the Chinese medicine group is not changed compared with mold group(p>0.05), where LH level reduce significantly compared with mold group (P<0.05).E2 level increase significantly compared with mold group (P<0.05). The light mirror descends growing follicecs and corpus luteums increased significantly in Chinese medicine group rats. yellow ovarian volume big,Atretic follicles reduced significantly. FSH of west medicine group rats reduced significantly compared with Chinese medicine group(P<0.05). whereas LH of Chinese medicine group rats reduced significantly compared with west medicine group(P<0.05).At other ways , Chinese medicine group is no significant different from west medicine.Part 2 The Clinical Study of YXBSF Treating Premature Ovarian FailureIn clinical research, Before treatment, the patients symptomswere very obvious, including prosopo-heating , sweating, circumgyration, cardiopalmus, restlessness and vagina drying.But all of the symptoms improved after treatment,and the significant deviation existed between before and after treatment .The menstrual cycle shortened contrast before,but it did not return norm . the endocrine level of POF patients is not significant different from pre-treatment(p>0.05) . FSH,LH level in the therapeutic group is not decreased notably after treatment compared with before(p>0.05), whereas E2 level increased significantly compared with pre-treatment (P<0.01). Endocrine level in blood serum has changed significantly compared with before in control group(p<0.05). After treatment endocrine level in two groups has no changed significantly group (P>0.05) .In treated group the excellent was 4,th effective was 10 and the ineffective was 6, the total effectiverate was 70%. In control group the excellent was 6, the effective was 9 and the ineffective was 5, the total effective rate was 75%. control group is no significant different from treated group(P>0.05).Conclusions1 The decreases of ovarian function, injury of organization pathology proved that the animal model of autoimmunity ovarian function decline had suceeefully founded.2 Changes of ovaries histology indicat that YXBSF can promote ovarian follicle and improve ovaries function.3 YXBSF can significantly reduce POF patients sweat, dry vagina, irritable irritability. thereby improving the quality of life for patients.4 YXBSF can improve POF patients endocrine levels, so the serum E2 levels increased significantly.
