Objective:To investigate the effect of perlecan on synthesis and secretion of TGF β1by cultured osteoblasts in vitro.Methods:Osteoblast-like cells were isolated from SD fetal rat calvarium and cultured, The cultured cells were identified using ALP staining and alizarin red staining. The experimental groups were divided into three sections randomly. Factors taken into consideration include the following:control:group,perlecan antibody group, and perlecan stimulation group. The synthesis and secretion of TGFβ1was examined in terms of immunohistochemical method and enzyme-linked immunosorbent assay(ELISA).Results:After we pretreated antibody of perlecan, the function that perlecan enhanced the proliferation of Osteoblast-like cells stimulated by TGFβ1was abolished, and TGFβ1concentration was decreased (P<0.05).Conclusion:perlecan could improve the effect of cultured osteoblasts synthesized and secreted TGFβ1in vitro.
