Part one:Establishment of rat model with leptin resistanceObjective: To establish the ideal rat model of leptin resistance.Methods: Female wistar rats were randomly divided into the model group (group M)and the control group (group N). The group M were treated with high-fat diets to induceleptin resistance while the group N were treated with normal diets. We measured rats’ bodyweight every week. Venous blood samples were obtained every two weeks for leptin andinsulin determination in. department of nuclear medicine in Tongji Hospital.Results: After4weeks of high-fat diets, the body weight and plasma leptin levelswere significantly higher than that in the control group (213.0±11.1vs230.1±7.8g, P<0.05;3.15±0.67vs8.87±1.27ng/ml, P<0.01, respectively); no significantly differences werefound in insulin levels (3.35±0.43vs3.48±0.45ng/ml, P=0.75).Conclusions: After4weeks of high-fat diets, leptin resistance was successfullyinduced.Part two: Effects of leptin resistance on implantation and endometrialreceptivity of ratsObjective: To investigate the negative effects of leptin resistance on implantation and endometrial receptivity.Methods: Female wistar rats were randomly divided into the model group (group M)and the control group (group N). The group M were treated with high-fat diets to induceleptin resistance while the group N were treated with normal diets. After modelsuccessfully building, all female rats were mated in estrus, the next morning, when spermwas found in vaginal smears, the first day of pregnancy was defined. Rats were sacrificedon day5,10of pregnancy. The pregnancy rate, average number of blastocyst, embryomorphology and blood supply were observed. The expressions of leukemia inhibitory factor(LIF) and integrin β3and their mRNA of the implantation sites were determined byimmunohistochemical method and PCR respectively.Results: Compared with the group N, pregnancy rate(93.3%,53.3%, P<0.01, averagenumber of blastocys(t12.18±4.63,5.12±1.67, P<0.01)and the levels of LIF(P<0.01)andintegrin β3(P<0.01) and their mRNA of the implantation sites were significantly reducedin the group M.Conclusions: Leptin resistance significantly reduces endometrial receptivity,5ofblastocyst, embryo morphology and blood supply were observed. The expressions ofleukemia inhibitory factor (LIF) and integrin β3and their mRNA of the implantation siteswere determined by immunohistochemical method and PCR respectively.Results: Compared with the group N, pregnancy rate(93.3%vs60.0%, P<0.01),average number of blastocyst(11.47±4.27vs5.07±1.32, P<0.01) and the levels of LIF(P<0.01) and integrin β3(P<0.01) and their mRNA of the implantation sites weresignificantly reduced in the group M. Compared with the group M, pregnancy rate(80.0%vs60%, P<0.05), average number of blastocys(t9.47±5.45vs5.07±1.32, P<0.05) and thelevels of LIF(P<0.05)and integrin β3(P<0.05)and their mRNA of the implantation siteswere significantly increased in the group C.Conclusions: Bushenhuatan Decoction could increase the expressions of LIF andintegrin β3in the implantation sites and improve poor endometrial receptivity andpregnancy rate.
