Objectives:In order to explore the long-chain transcription of non-coding RNA HOXantisense RNA (HOTAIR mRNA) and renal cell carcinoma, the development of therelationship between the detection of non-coding RNA HOTAIR expressiondifferences between normal renal tissue near the renal clear cell carcinoma and cancer,and further explore its relationship with renal cell carcinoma each clinical stage,pathological grade. By constructing shRNA vectors to study the non-interferencesilencing of the gene encoding HOTAIR786-0clear cell renal cell carcinoma growth,apoptosis, invasion and migration affect the biological behavior, diagnosis andtreatment of kidney cancer provides new methods and ideas.Methods:Collected renal clear cell carcinoma tissue and adjacent normal kidney tissuewhich diagnosed by urology laparoscopic postoperative pathological between August2013to April2014in the First Affiliated Hospital of Nanchang University.Extracted the total RNA of renal clear cell carcinoma tissue and adjacent normalkidney tissues by TRIZOL. Detected the HOTAIR expression in renal clear cellcarcinoma tissue and adjacent normal kidney tissues by real-time PCR. Combinedclinical stages and pathological grading, and carries on the statistical analysis. Usedgene silencing methods, constructed shRNA carrier, researched the effect of silencingthe non-coding RNA HOTAIR in renal clear cell carcinoma786-0cells growing:test cell proliferation by CCK-8, detect cell invasion by transwell test, and detect cellapoptosis by FCM.Results:1、 The expression of HOTAIR in renal clear cell carcinoma was12.36±1.61, whileits adjacent normal kidney tissues was3.01±0.45, the expression of non-codingRNA HOTAIR in renal clear cell carcinoma was significantly higher than that inadjacent normal kidney tissues (P <0.05).2、 The expression of non-coding RNA HOTAIR in renal clear cell carcinoma were significantly increased from high to low differentiation group and clinical stage Ito clinical stage III, with significant differences between groups (P <0.05),suggesting the high expression of HOTAIR in renal clear cell carcinomaassociated with tumor malignance, while has no statistical difference in age andsex (P>0.05).3、 After silencing the non-coding RNA HOTAIR, the proliferation of renal clear cellcarcinoma786-0cells lowed significantly and cell invasion ability declinedobviously, while cell apoptosis increase obviously.Conclusion:1In renal clear cell carcinoma, expression of HOTAIR significantly upregulated,suggesting that non-coding RNA HOTAIR renal cell carcinoma may be associatedwith the occurrence and development related to the detection HOTAIR expressionlevels may contribute to the diagnosis of renal cell carcinoma.2HOTAIR expression levels and renal cell carcinoma was positively correlatedwith clinical stage, perhaps as a diagnosis of renal clear cell carcinoma or determinenew indicators renal cell carcinoma prognosis.3In vitro, experiments found that silencing the expression of endogenousHOTAIR can effectively inhibit the proliferation of renal cancer786-0, reducing itsinvasiveness and promote apoptosis of cancer cells.4Non-coding RNA HOTAIR in renal clear cell carcinoma may play a role incancer-promoting genes down-regulated expression, may become a new means oftreating kidney cancer.
