Objective: To analyze the lnc RNA NR-028414 expression and association with occurrence and development of clinical and clinicopathological parameters in renal clear cell carcinoma and normal renal tissue.To investigate the clinical value of lnc RNA NR-028414 in renal cell carcinoma diagnosis;Using the method of gene silencing to synthesize lnc RNA NR-028414 gene-specific si RNA.To research the impact of lnc RNA NR-028414 inhibition on the proliferation,apoptosis,invasion and migration of human renal clear cancer cells A498,and to explore its possibility as a new target of renal clear cell carcinoma.Methods: Collected 283 cases of renal clear cell carcinoma tissue(without chemotherapy or radiotherapy)and 30 cases of normal kidney tissue which treated with surgical treatment and diagnosed by postoperative pathologic diagnosis between January 2011 to January 2015 in the Department of Urology,Affiliated Hospital of Qingdao University.The total RNA of renal clear cell carcinoma tissues and normal renal tissues was extracted by TRIZOL method and reverse transcribed.To determine the expression of lnc RNA NR-028414 in 283 cases of renal clear cell carcinoma and 30 cases of normal renal tissue by real-time PCR.To analyze the relationship between the expression of the lnc RNA NR-028414 and the clinical data of patients combined with Fuhrman classification and clinical stage,using SPSS 19.0 software for statistical analysis.(2)The chemically synthesized lnc RNA NR-028414 gene specific si RNAs was transfected into renal cancer cells A498 using the method of loss of gene function,reduced expression of lnc RNA NR-028414.The proliferation of cells was detected by cell proliferation assay(CCK8).Apoptosis was detected by cell apoptosis(flow cytometry).Transwell cell invasion assay was used to detect the invasive ability of cells.Cell scratch test was used to detect cell migration.Results:(1)Real-time PCR show that high levels of lnc RNA NR-028414 expression were observed in renal clear cell carcinoma tissue specimens,and low lnc RNA NR-028414 expression in normal renal tissue samples.(P<0.001);The lnc RNA NR-028414 expression of Fuhrman in the high level group was significantly higher than that of low level group(P<0.05),the lnc RNA NR-028414 expression of clinical advanced stage is higher than that of the early group(P<0.05),indicating that lnc RNA NR-028414 expression in renal clear cell carcinoma may be related to tumor malignancy.However,no obvious relationship between lnc RNA NR-028414 with gender,age(P>0.05)(2)The expression of lnc RNA NR-028414 was significantly decreased after transfection with lnc RNA NR-028414-si RNA transfected with renal clear cell line A498(P<0.05).The proliferation of renal clear cell carcinoma cells was significantly decreased(P<0.05)and the apoptosis was increased(P<0.05).The cell invasion of lnc RNA NR-028414 was decreased(P<0.05)and the cell migration ability was decreased(P<0.05).Conclusion:(1)In renal clear cell carcinoma,lnc RNA NR-028414 was overexpressed,indicating that lnc RNA NR-028414 may be related to the development of renal clear cell carcinoma.The expression of lnc RNA NR-028414 was positively correlated with Fuhrman grade and clinical stage of renal clear cell carcinoma,indicating that lnc RNA NR-028414 may be related to the degree of malignancy.The lnc RNA NR-028414 could be a new marker for the diagnosis of renal clear cell carcinoma.(2)Reducing the expression of lnc RNA NR-028414,can inhibit the proliferation of renal clear cell carcinoma cell line A498,increase apoptosis,reduce cell invasion and migration ability,indicating that lnc RNA NR-028414 can play a role in the occurrence and development of renal clear cell carcinoma as an oncogene,which indicated that it can be designed for a specific target treatment for lnc RNA NR-028414.
