Objective: Establish a melamine and cyanuric acid-induced kidney stone model which is noabnormal kidney function,then explore the possible intervention effect of Clerodendranthusspicatus extract and Rosmarinic acid on urinary and reproductive systems.Methods:①50ICR mice(♂) were randomly divided into five groups: normal controlgroup(qod), model group(qod), low RA group(qod), high RA group(qod), Uralyt-Ugroup(qod). The normal control group was given with NS by oral gavage and the otherswere given with the suspension of melamine-cyanuric acid by oral gavage at a dosage of25/25mg/(kg·d) at8a.m. every other day. And then, NS, NS, low RA(50mg/kg), highRA(100mg/kg) and Uralyt-U(2.5g/kg)were respectively given by oral gavage at16p.m.daily. All mice were killed after30days and serum, kidneys and testis were collected.②40ICR mice(♂) were randomly divided into four groups: model group(qd), RA group(qd),Orthosiphon group(qd), Uralyt-U group(qd). Each group were given with the suspension ofmelamine-cyanuric acid by oral gavage at a dosage of25/25mg/(kg·d) at8a.m. continuously;And then, NS, RA(50mg/kg), Orthosiphon stamineus extract (1g crude drug/ml) andUralyt-U(2.5g/kg)were respectively given by oral gavage at16p.m. every day. All micewere killed after15days and serum, kidneys and testis were collected.③Analyses included arange of body weight,water consumption, Cr, BUN, UA, ALT, AST, MDA, SOD, renal andtestis histopathology and melamine content in the kidneys.Results:①There were not significant different between each group in body weigh, serumbiochemical, SOD and MDA after30-days exposure of a mixture of melamine/cyanuric acid(25/25mg/(kg·d))every other day. Compared with normal control group(qod) and Uralyt-Ugroup(qod), some yellow substance deposited in renal tubular with epithelial cell damage,fall off and appotosis and reduce the number of sperm cells in model group(qod). Themeasured results of melamine in the kidneys of mice:low RA group(qod)>model group(qod) >high RA group(qod)>Uralyt-U group(qod)>control group(qod).②There were notsignificant different between each group in serum biochemical15-days exposure of a mixtureof melamine/cyanuric acid (25/25mg/(kg·d)) continuously. There were lots of melaminecrystals in renal tubular with epithelial cell damage, fall off and appotosis and spermatogeniccellseries necrosis and fall off and the number of interstitial cell decrease in model group (qd).The measured results of melamine in the kidneys of mice:RA group(qd)>model group(qd)>Uralyt-U group(qd)>Orthosiphon group(qd).Conclusion: Orthosiphon stamineus extract play a renoprotective role in melamine andcyanuric acid-induced kidney stone in mice. Rosmarinic acid may also have same effect.
