ObjectiveDiscuss whether there are differences between hemophagocytic syndromepatients and normal human bone marrow CD34+cells’ quality, research the effectand characteristics of interferon gamma differentiation to the bone marrow, andobserve whether improve colony stimulating factor can restrain the inhibition ofinterferon gamma on bone marrow. Clarify the pathophysiological mechanisms ofIFN-gamma mediated HPS blood cell reduction, to guide the clinical medication,explore new methods to provide theoretical basis for treatment.Materials and MethodsBone marrow taken from16cases diagnosed by clinical and laboratoryexamination of HPS patients; taken from11cases of normal bone marrow transplantdonors. The HPS patients were initially treated patients, the groups before samplingwere not system used immunosuppressant, retinoic acid and chemotherapy drug.Bone marrow were provided by the Department of Hematology of Beijing FriendshipHospital, taken after the agreement of patients, the protocol was approved by thehospital ethics committee. Separation and purification bone marrow CD34+hematopoietic stem cells by immunomagnetic beads method. SCF, IL3, GM-CSF,G-CSF, EPO colony stimulating factor on differentiation of bone marrow stem cells,the colony forming units (CFU) to evaluate the formation of bone marrowdifferentiation. Added different concentration gradient of interferon gamma in thesystem, observation the bone marrow colony formation the situation. Increase in the minimum concentrations interferon gamma inhibition of bone marrow in colonystimulating factor concentration to observe the bone marrow growth situation.Results1. This study demonstrated no difference in bone marrow CD34+cells comparedwith normal quality, P>0.05.2. The degree of inhibition of interferon gamma in different concentration on bonemarrow is different, when interferon gamma concentration increased to more than10ng/ml, the experimental group colony number difference compared to thecontrol group without the addition of interferon gamma, P <0.05.3. To increase the training system of colony stimulating factors concentration can bemade against the bone marrow suppression of interferon gamma.Conclusions1There is no difference of bone marrow’s quality between HPS patients and normal.2Interferon gamma may inhibit bone marrow’s growth.3Improve the colony stimulating factor concentration inhibition may againstinterferon gamma production. |
