Objective: Testicular torsion is one of the infantile scrotum surgical emergency,it is difficult to diagnosis early, so it been delayed treatment and has high medical risk,the sequelae as testicular dysfunction after operation has been reports, but not hasspecial treatment for it now. Our study in order to investigate the protective effects ofvitamin E in the rat’s unilateral testicular torsion after resetting, provide theexperimental basis for clinical treatment of testicular torsion.Method: The36Sprague-Dawley rats-health, bilateral testicular development-were randomly divided into3groups,with12rats in each group, respectively for theshame operation group (S group), torsion/reset group (T/R group) and vitamin Etreatment group (V group). S group optional flip a few times on the left side of thetesticles, observe8h after excision of testicular; T/R group of the left testicular torsionmodel is established according to the Turner, it been fixed on2h then restoration toobserve6h after be resection. V group rats is establishment the left testicular torsionmodel, intramuscular injection vitamin E200mg/kg before0.5h that it be restoration,after observation6h then to removed the left testis. Testes specimens respectivelymaking tissue homogenate, used to determine of the tissue SOD、MDA、cytokine andTUNEL apoptotic cell index be calculation.All the experiment data to adopt x±s, by SPSS19.0statistical analysis, usingthe single factor analysis of variance and F test. Variance together when using LSDtest and variance is used when not neat Dunneltt ’s T3test, the results with P <0.05for statistical significance.Result:1.By the tissue homogenate SOD and MDA content determination.compared with S group, the T/R group and V group all obviously that the SODcontent decreases and MDA content increased (P <0.001);And compared with theT/R group, SOD activity in the value of V group to rise(P=0.007) and reduce MDAcontent has obvious change(P=0.015<0.05).2. Cytokines in each specimen (IL-1、IL-6、TNF-alpha) content comparison found that compared with S group, the rest of the two groups of cytokine levels haveincreased significantly (P <0.001); And V compared with T/R group, the content ofcytokines increase less(IL-1: P=0.003; IL-6: P=0.04; TNF-α: P=0.03).3. Then observed the changes of each specimen by HE staining, the rankstatistics of organizational structure found that the organizational structure of the Vand T/R group compared with the S group, the organizational structure of the formerslevel is highly, the damage is seriously (P <0.001); and a comparison between thetwo, before T/R structure damage more seriously(P=0.006<0.05).4. By the TUNEL method marked statistical analysis of the apoptosis cells indexof testicular tissue, compared with S group, the rest of two groups of apoptosis indexincreased significantly (P <0.001), compared to the V and T/R group, the apoptosisindex of V group was obviously decreased (P=0.012<0.05).Conclusion: Vitamin E by increasing SOD activity and reducing MDA andinhibit the production of cytokines, to improving the tissue structure of testicular andreduced the cell apoptosis, to protective the testicular tissue after torsion and resetting,against the testicular ischemia reperfusion injury.
