Surface plasmon resonance (SPR) is optical sensing technique. It has beenwidely used in various fields, such as bimolecular interaction, clinical diagnosis andenvironmental monitoring, etc. But its application in Traditional Chinese Medicinehad never been researched. Chronic skin ulcer is a clinical refractory disease. Theulcer wound repair is complex. A variety of cells and tissue factors are involved inthis process. In order to explore the effects of each factor in the wound healing, it isnecessary to detect the content of these factors during the process. There are someimmunological detection methods have been used in wound repair. But they all getsome disadvantages, such as complicated operation, consuming time and samples,and poor repeatability. These disadvantages not only lead to the single sample’sreal-time detection is inconvenient, but also result in that a large size of samples areneeded in some quantitative detection methods. Because of these disadvantages, theimmunological methods are unsuitable for the clinical testing. SPR can givelabel-free, highly sensitive and real-time monitoring with a small amount of sample.So it is possible for SPR to use in chronic skin ulcer clinical detection.The long-range SPR (LRSPR), as its higher performance, has attracted moreand more attention. Because of its characteristic of the structure limits its detectionrange, the practical application has been affected. We proposed a new symmetricalstructure LRSPR based on MgF2-Au-MgF2film and achieved the instrumentation.Then we tested the performance of the LRSPR biological sensor. The results showedthat the sensitivity was1.22×105pixels/RIU and the system resolution of themachine was6.1×10-7RIU.After achieving to make the symmetric structure LRSPR biosensor intoinstrument, we used it to detective the clinical secretion of chronic skin ulcer. It isan exploratory research. As the secretion of chronic skin ulcer is clinical sample, wetried to establish the inspection process. The process included all the steps, such assampling methods, processing, and the quantitative detection. What’s more, wedetected some related factors. The results showed that the biosensor hadsuccessfully detected the factors’ change in the patient samples at different period. Itcould be concluded that the symmetric structure LRSPR biosensor, the instrumentused in this thesis, could be used in the clinical detection to detective the secretionof chronic skin ulcer.
