Objective:This experiment uses the chinese medicine surgical preparation new square skin sticky(after the improvement of the skin sticky square)as the research object,through animal model research,further expounds the mechanism of the effect of the new square skin adhesion to treat chronic skin ulcer under the guidance of "skin treatment",and verifies its clinically related function.Methods: The experimental subjects of the experiment a total of 24 SD rats,adaptive feeding for 1 week,with 50% ice acetic acid solution coated face(1 time /d,a total of 7d),the formation of a defective skin ulcer model,24 rats randomly divided into model groups,thin skin sticky group,elephant skin group,borax group,1 time per day to give medicine,will be uniform external drugs Scatter in the rat back ulcer,after 21 days of administration,the execution of animals,take local ulcer tissue for immunofluorescent detection of new blood vessel-related expression factor CD34 content,immuno-group chemical staining detection collagen-I and collagen-IV expression,combined with the healing of the rat skin ulcer,to investigate the role of thin skin sticky to promote the healing of chronic skin ulcers.Results: Immunofluorescent determination CD34 expression: the skin adhesion group can see a strong positive CD34 fluorescence signal,and form a tube cavity structure;Immunofluorescent determination collogen I fluorescence intensity: model group: positive fluorescent signal is strong,fluorescent staining seen collagen I arrangement disorder;skin adhesion group: positive fluorescence signal strong,fluorescence dyeing seen collagen I arrangement consistency is better;elephant skin group: positive fluorescence signal is weak,fluorescence stained collagen I.better arrangement;borax group : Positive fluorescence signal is weak,fluorescent staining seen collagen I arrangement disorder.Immunofluorescent determination of the fluorescence intensity of collagen IV: model group: positive fluorescent signal is weak,fluorescent staining saw the organizational structure disorder,the formation of a small number of tube cavity structure,no epidermal tissue base membrane formation;Elephant skin group: positive fluorescent signal is strong,fluorescent staining shows a large number of tube cavity structure,visible epidermal tissue base membrane formation,but not continuous;Immunofluorescent determination TGF-beta 1: Positive fluorescence signals were weak in each group,and there was no significant difference.The area of the creation surface: In 7,14,21(days)after the prescription of the wound,there were significant differences between the area of the thin skin sticky group,the wounding area of the elephant skin group and the model group,and the thin skin sticky group and the elephant skin group were significantly better than the model group;(P<0.01);There was no significant difference between the area of the model group's creation surface and the borax set(P>05);Create face color: 7,14,21(days)after the wound ingestised administration,there are significant differences between the skin sticky group and the model group(P<0.05);There was no significant difference between the elephant skin group and the borax group compared with the model group(P>05);Wounded secretions and odors: 7,14,21(days)after the wound ingesting wasgiven,there was no significant difference between the thin skin adhesion,the elephant skin group,and the borax group compared with the model group(P>05);Fresh-faced granulolate tissue: 7,14,21(days)after the wound ingesting wasgiven,there were significant differences between the skin adhesion group and the model group(P<0.05);There was no significant difference between the elephant skin group and the borax group compared with the model group(P>05);Conclusion: This experiment showed that skin viscose has a relatively good healing effect on chronic skin ulcers in rats,while immunofluorescent detection of CD34,collagen I,IV expression increased.Single taste use of elephant skin and borax,not only from an intuitive point of view can not effectively promote the healing of rat ulcer wound,in the micro-angle also suggested that the two-flavored drug alone use of rat ulcer wound-related factors expression is not apparent.But the combination of twoflavored drugs can produce good results both intuitively and microscopicly.
