[Objective] To explore the mechanism optimize treating mechanism induced by Lang-du extrct(LDE) by investigating the effect on apoptosis, cellcycle and the expression of bcl-2, bax, caspase-9mRNA, PCNA, TSP-1in Lewis lung carcinoma.[Methods] Different concentration of LDE were used to assesss its effect on mouse transplanted Lewis lung carcinoma and its influence on tumor growth in vivo and in vitro including cell cycle, apoptosis rate, bcl-2mRNA, bax mRNA, caspase-9mRNA, PCNA, TSP-1, Anti-oxydent capacity.[Results](1) The model mice were successfully builded, which laid a experimental basis for the further research on mechanism of LDE action.(2) The in vitro experiment shows that the lewis cells, after intervened by different concentration of LDE, were block in G0/G1phase, the apoptosis rate significantly rised, the same to the expression of bax, bcl-2, caspase-9mRNA, PCNA, TSP-1, compared to control group, the diffenrence were significance (P<0.05).(3) The in vivo study indicate that the mouse transplanted Lewis lung carcinoma, after intervened by LDE, were block in G0/G1phase, the apoptosis rate significantly rised, the same to the expression of bax, bcl-2, caspase-9mRNA, PCNA, TSP-1and antioxidant capacity, compared to control group the diffenrence were significance(P<0.05).[Conclusions](1) LDE of different concentration can induce apoptosis and depress the proliferation in different dgree.(2) LDE can induce apoptosis and depress the proliferation in different degree to the mouse transplanted Lewis lung carcinoma, as the same to antioxidant capacity. |
