Part Ⅰ The EPAS1and VEGF expression in endometrial carcinomaObjective To investigate the expressions of endothelia PAS domain protein1(EPAS1),vascular endothelial growth factor(VEGF) and the relationships betweenthem in human endometrial carcinoma.Methods In46cases of resected specimen of endometrial carcinoma and thecorresponding46cases of normal endometrial tissue,RT-PCR was used to detect theexpression levels of EPAS1and VEGF mRNAs,western blotting was used toexamine the expression levels of EPAS1and VEGF proteins respectively.Results Higher expressions of EPAS1and VEGF were detected in endometrialcarcinoma than in normal endometrial tissue, for EPAS1both at mRNA and proteinlevel(t=4.5871,p=0.0048;t=19.4877,p=0.0001),the same as the mRNA and proteinlevel of VEGF (t=7.0495,p=0.0001;t=98.4145,p=0.0001).Conclusions Both EPAS1and VEGF were overexpressed in endometrial carcinoma.Part Ⅱ The clinical significance of EPAS1and VEGF expressionin endometrial carcinomaObjective To investigate the expressions of EPAS1, VEGF and microvesseldensity(MVD) as well as the relationships among them,and the relationships to theclinicopathologic features of human endometrial carcinoma.Methods In46cases of resected specimen of endometrial carcinoma and thecorresponding46cases of normal endometrial tissue, immunohistochemical SPmethod was used to examine the expression levels and distributions of EPAS1andVEGF proteins respectively.MVD regarded as a marker of angiogenesis was exploredalso.The correlations among them and their relationships to the clinicopathologiccharacteristics of human endometrial carcinoma were analyzed.Results Higher expressions of EPAS1,VEGF and MVD were detected in endometrial carcinoma than in normal endometrial tissue.Furthermore, significantcorrelations were observed among them(between EPAS1and VEGF,r=0.7358,P=0.0041; between VEGF and MVD, r=0.8871,P=0.0001; between EPAS1and MVD, r=0.7360,P=0.0003),the positive expressions of EPAS1and VEGF wereclosely related with FIGO staging and infiltration of endometrial carcinoma.Conclusions Both EPAS1and VEGF were overexpressed in endometrial carcinoma,and the expression of EPAS1significantly direct correlated with that of VEGF andthe value of MVD. EPAS1may be involved in the angiogenesis of endometrialcarcinoma by upregulating the expression of VEGF, and play an important role inthe carcinogenesis and aggression in endometrial carcinoma....
