Effect Of Okam On Cyokine Levels In Bronchoalveolar Lavage Fluid And Lung Tissues Lung Tissue Of Asthmatic Mice

Objective This paper aims to observe the expression of IL-17, IL-6, HB-EGF in bronchoalveolar lavage fluid and lung tissues in mouse with asthma. How okam effect the expressions was also researched to explore the possible regulatory mechanisms.Methods Thirty-two SPF grade KUNMING STRAIN mice were randomly divided into four groups:control group, asthma group, budesonide-treated group, okam-treated group, with eight mice in each group. The asthmatic mice model was produced by combined injection and aerosol inhalation OVA. The histological changes in the bronchi and alveoli were observed under microscope by hematoxilin and eosin(HE) staining and the asthma model was evaluated by histological analysis of lung tissues. The levels of IL-17, IL-6, HB-EGF in BALF were assessed by enzyme linked immunosorbent assay(ELISA). Levels of cyokines in lung tissues were assessed by immunohistochemistry staining.Result:The changes in lung tissues in the okam and budesonide groups were compared with those of the asthma group, showing significant differences between the okam and budesonide group. Compared with control group, IL-17, IL-6and HB-EGF expressions were significantly enhanced in lung tissues of mice in asthma group (all p<0.05), Compared with asthma group,IL-17,IL-6and HB-EGF protein levels were significantly decreased in lung tissues of mice in okam or budesonide treatment groups; There were significant difference between okam or budesonide treatment group for IL-17(P<0.05);IL-6HB-EGF level’s difference isn’t significant(P>0.05). Immunohistochemical staining showed that IL-17, IL-6, HB-EGF have a positive expression in the airway epithelium of mice. For IL-17, IL-6, HB-EGF, the positive index in budesonide and okam treated groups is obviously lower than that in asthma group (all P<0.05). However, the positive index for these three cyokines in okam-treated group was obviously higher than that in budesonide-treated group (all P<0.05).Conclusion The expression of IL-17, IL-6, HB-EGF in BALF and lung tissues of asthmatic mice were increased, which demonstrated that all of them took a part in the process of asthma inflammatory reaction.To a certain extent, Okam can improve the asthmatic airway inflammation and remodeling in mice via inhibiting the expression of IL-17, IL-6,HB-EGF.
