BackgroundLung cancer has been one of the most serious diseases threatening human health, and more than1million people die from it around the word every year. The morbidity of lung cancer is continuously increasing, and has become the highest rates of malignant tumors in our country. Non-small cell lung cancer accounts for about80%of lung cancer, while the5-year survival rate was15%only.70%of the patients with lung cancer at the time of diagnosis has been too late to have the opportunity to surgery and lack of effective treatment because of it has been locally advanced or distant metastasis (Ⅲb/Ⅳ period), and the5-year survival rate less than15%. While if patients of lung cancer can be diagnosis and treatment at I a period, the5-year survival rate can reach80%. Therefore, research of occurrence and development mechanism of lung cancer has important guiding significance for the early diagnosis, treatment and stratify of prognosis and efficacy of tumor.ObjectiveTo investigate the expression and clinical significance of HMGA1, CD147and MMP-2in non-small cell lung cancer (NSCLC), and analysis the relationship of the expression and biological behaviour of NSCLC, so as to provide references for clinical diagnosis, clinical treatment and prognosis estimation.MethodSurgically resected95cases of NSCLC tissue with complete clinical informations and pathological confirmed specimens, and25cases of adjacent normal lung tissue (located at least10cm away from the tumor margin) were collected from shanghai Yodak Cardio-Thoracic Hospital and Fu Dan Clinical Pathology Diagnosis Center. All the patients received neither radiocherapy nor chemocherapy before the surgery. Expressions of HMGA1, CD147and MMP-2in NSCLC tissues and adjacent normal lung tissues was determined by immunohistochemistry assay (S-P method). All the results were statistically analyzed with SPSS19.0software package.Results(1) The expression of HMGA1protein in NSCLC was63.16%(60/95), which was much higher than8%(2/25) in normal lung tissue (P≤0.05). The protein expression of HMGA1of adenocarcinoma and squamous cell carcinoma of lung was significantly correlated with lymph node metastasis and TNM stages (P<0.05), but not with ages, gender, smoking history, histological subtypes and tissue differentiation degree (P>0.05).(2) The expression of CD147protein in NSCLC was80%(76/95), which was much higher than12%(3/25) in normal lung tissue (P≤0.05). The protein expression of CD147of adenocarcinoma and squamous cell carcinoma of lung was significantly correlated with lymph node metastasis and TNM stages (P≤0.05), but not with ages, gender, smoking history, histological subtypes and tissue differentiation degree (P>0.05).(3) The expression of MMP-2protein in NSCLC was76.84%(73/95), which was much higher than4%(1/25) in normal lung tissue (P≤0.05). The protein expression of MMP-2of adenocarcinoma and squamous cell carcinoma of lung was significantly correlated with lymph node metastasis and TNM stages (P≤0.05), but not with ages, gender, smoking history, histological subtypes and tissue differentiation degree (P>0.05).(4) Using Spearman correlation test to analyze the correlation among HMGA1, CD147and MMP-2expression in NSCLC tissues. Expressions of HMGA1and MMP-2in NSCLC tissues showed positive correlation (rs=0.786,P≤0.05). Expressions of HMGA1and CD147in NSCLC tissues showed positive correlation.(rs=0.328, P≤0.05)Conclusions(1) The expression of HMGA1, CD147and MMP-2in NSCLC tissues were significantly correlated with lymph node metastasis and TNM stages, while not correlated with ages, gender, smoking history, histological subtypes and tissue differentiation degree.(2) The expression of HMGA1and CD147correlated to MMP-2positively, and the expression of HMGA1correlated to CD147positively. Combinational examination of HMGA1, CD147and MMP-2may have a collaborative or mutual regulation role in the process of occurrence and development in the NSCLC.
