Research backgroundLeprosy is a chronic diseases which caused by the mycobacterium, skin and peripheral nerve involvement in. Human beings are the main host of the mycobacterium and the source of infection. Untreated leprosy patients(mainly the multibacillary leprosy patients) can infect through the respiratory tract and skin.the leprosy patients with different degrees of cellular immune deficiency. The precise mechanisms of cells immunodeficiency is still unknown.Studies have shown that cells of different subsets of T helper cells (Th) dysfunction plays an important role in the pathogenesis of leprosy development.And Thl, Th2, Th17and Treg imbalance could lead to leprosy patients with immune dysfunction trigger and exacerbate the patient’s condition changes.Different type of leprosy performance is the result of numerous cytokines synergistic effects, Th cell subsets in the release of many cytokines play network adjustment effect in various stages of leprosy. different cytokine plays a complex disease, the development and transfer process to promote and (or) inhibition.Moderating Effect of different subsets of Th cell cytokine network, may help us to clarify the incidence of leprosy development processand and be benefit to leprosy vaccine development.aslo lay the foundation for further study of the role played by the different Th cell subsets in the development of the incidence of leprosy pathogenesis.methodsSpecimens from dermatological department and clinical laboratory of the second affiliated hospital of kunming medical college, the yunnan centers for disease control and prevention, the xishan district centers for disease control and prevention in kunming,the guandu district centers for disease control and prevention in kunming.Which has25cases cured leprosy patients, leprosy patients XianZheng15cases,20cases of normal control group. Using flow cytometry respectively to detect the four groups of CD4+CD25+CD127lowTreg accounts for the percentage of CD4+, and detection of IFN-gamma, IL-4, IL-17SPSS19.0software package for data for statistical analysis.Objectives(1)By comparing the current disease leprosy patients, cured lepers percentage of patients with CD4+CD25+CD127Iow accounted for CD4+, and analyze the relationship between the number of Treg cells and the clinical manifestations of leprosy.(2) Systems analysis between leprosy patients and normal controls, that cure group, current disease group, the more bacteria disease group, and the less disease bacteria group, cure multiple bacteria group, cure less bacteria group between bacteria and normal IFN-γ, IL-4, IL-17, under different clinical manifestations, the relationship between leprosy patients Thl, Th2, Th17cells and Treg cells.Results1, current and control groups, cure and control groups percentage of peripheral blood CD4+CD25+CD127lowTreg in to CD4+significant difference (P<0.05).2, cure bacterium group and with less strain group of CD4+CD25+CD127low Treg in peripheral blood, have significant differences, cure bacterium group and Disease bacterium group of CD4+CD25+CD127l0W Treg in peripheral blood, have significant differences. Disease less strain group and cure less strain group of CD4+CD25+CD127low Treg in peripheral blood, have significant differences. Disease less strain group and Disease bacterium group of CD4+CD25+CD127low Treg in peripheral blood, have significant differences.3, current and control groups, cure and control groups IFN-γ, IL-4, IL-17, three kinds of cytokines in significant difference (P<0.05).current and cure groupsin significant difference (P<0.05).current and control groups, cure and control groups IFN-γ/IL-4in significant difference,current and cure groupsin significant difference (P<0.05).4, Disease bacterium group and with less strain group of IFN-γ in peripheral blood, have significant differences.Disease bacterium group and with less strain group of IL-17in peripheral blood, have significant differences,aslo IFN-γ/IL-4between Disease bacterium group and with less strain group.5, Disease bacterium group of IFN-γ/IL-4and CD4+CD25+CD127low T/CD4+T negative correlation, less strain group of IFN-Y in peripheral blood/IL-4and CD4+CD25+CD127low T/CD4+T were positively correlated, cure bacterial group of IFN-Y in peripheral blood/IL-4and CD4+CD25+CD127low T/CD4+T negative correlation.6, Current less disease bacteria group IL-17and CD4+CD25+CD127lov T negative correlation.Conclusions1, The number of Treg in the process of reduction and increase may be closely related to the evolution of leprosy type.Treg can inhibit the inflammatory response to IL-17, suggesting that early stage disease, Treg can balance the CK in favor of immunity against that leprae, and with chronic stage, Treg cells they may inhibit cellular immunity, delayed healing of the disease, and also susceptibility.2,suggesting that IL-4, IFN-γ and IL-17interact with Treg cell in leprosy.3,suggesting that has cured leprosy patients while clinical symptoms disappeared completely, but not the immune function.
