[Objective]To verify different protein expression detected in the normal group, patients with SLE activity groups, and the patients with stable SLE group, ELISA, Western blot and immunohistochemical technique were apllicated in serum and skin lesions respectively. In order to set a valuable diagnostic indicators, SLE biological marker was detected.[Methods]1.ELISA method was applied to detect different expression of neutrophils defense element1,3, C3, anda-1anti-pancreatic chymotrypsin in serum of SLE stable group, SLE activity group and normal control group respectively.2.The application of immunohistochemical technique was detected Heat shock70-kDa protein, Heat shock90-kDa proteinl, SCCA1, SCCA2, Keratin2e, Keratin6, Involucrin, Keratinl expression in skin lesions.3.Western blotting can be used to further verify the valuable proteins of skin above the experiments.[Results]1. Heat shock90-kDa was expressed, which colour was darker at the lesion corneous layer and Heat shock90-kDa contains mild expression at normal group corneous layer and the base layer of cutin formed cell cytoplasm.Heat shock70-kDa was showed no difference in lesion group and normal group of corneous layer and base layer.SCCAl is expressed at full-thickness of the lesion skin, which colour was dyed deeply, however,SCCAl has mild expression at base layer and corneous layer of the normal group. SCCA2has only mild colored expression at corneous layer of the normal group, which mainly colored tan in the stratum corneum of the skin lesions, spine layer and base layer of cutin cell cytoplasm. Keratinl, pale yellow, detected in skin lesions mainly concentrated at upper-middle-classof the epidermis.Full-thickness skin cells were colored tan in the normal group, which are expressed slightly stronger than SLE lesion group.Keratin6at base layer are scattered colored or not in normal group,but which colored at the keratin layer and base layer in the skin lesions group.Keratin2e colored mainly is concentrated at the upper epidermis of skin lesions, which is tan and colored pale yellow at upper epidermisof the normal group that is slightly weaker than that in SLE lesion group.2.The application of ELISA method is detected the HNP1-3, and alpha-1-AT in the group of SLE activity, stable SLE group, and normal serum group. There is no statistical difference between them (P>0.05), the C3expressed lower in activity group compared with the normal group.3.Western blotting test showed that Cytokeractin6, Hsp90and SCCA expressed in normal skin group and skin lesionsgroup, in which Cytokeractin6and Hsp90in skin lesions are expressed significantly higher than in normal groups.[Conclusions]1. There are some different proteins expresssion between SLE lesiongroup and normal skin group. The expression of Involucrin proteins, Keratin6,Keratin2e,Heat shock90-kDa proteinl, SCCA1, SCCA2increased in the lesion of SLE group, but the expression of Keratinl decreased. The high expression of Keratin2e、Keratin6、 Involucrin proteins and the low expressionof Keratinl in lesion group perhaps relates to keratinocyte activation, cytokine release, keratinocyte abnormal differentiation and the forming of SLE lesion.2.Complement C3maybe has significant clinical value of diagnosis of SLE.3.The alpha-1,anti-pancreatic chymotrypsin and neutrophils defense element1,3in the role of SLE remains to be further defined.
