Effect Of Compound Rhizoma Curcumae Power On Expression Of CyclinA And P21in Endometriosis Patient

Endometriosis is a common disease, seriously affecting young women’s the health and quality of life. The pathogenesis is complex, and it can contribute to extensive lesions. There are various theories of the etiology, such as the doctrine of endometrial implantation, lymphatic and venous spread the doctrine, the doctrine of the body cavity metaplasia, induction theory, genetics and immune regulation theory and so on. But these theories can not explain similar clinical behavior of malignant tumors of endometriosis, such as planting, invasion and distant metastasis. In recent years, many researchers consider disease progression and etiology of endometriosis from the perspective of cancer pathogenesis. It has been demonstrated that the incidence of malignant tumors is related to abnormal cell cycle, then there are many domestic and foreign scholars who have done a preliminary study on relationship between cell cycle and endometriosis. Thus can be speculated that incidence of endometriosis may be associated with abnormal expression and proliferation of certain cell cycle proteins and so on. We select cyclinA and P21as indicators, observe the expression of them in ectopic and eutopic endometrial tissue in patients and difference between groups, to study their roles and significance in the occurrence and development procession of endometriosis. Professor Cao Baoli developed the representatives prescriptions of treatment of endometriosis, compound Rhizoma Curcuma powder, based on years of clinical experience and the basic condition in Chinese medicine,"Kidney Blood".We examined the affection of compound Rhizoma Curcuma powder on expression of cyclinA and P21in eutopic and ectopic endometriosis in patients, thereby providing comprehensive study of the experimental data for the clinical treatment of endometriosis.Objective:1.To observe the expression of cyclinA and P21in eutopic and ectopic endometriosis in patients, thereby exploring their roles and clinical significance in occurrence and development of endometriosis.2.To study the effect of compound Rhizoma Curcuma powder on expression of CyclinA and P21in eutopic and ectopic endometria on endometriosis patients, study the mechanism and provide implications for administrating compound Rhizoma Curcuma powder in clinic. Method:1.Select the method of case-control studies,CyclinA and P21protein expression in patients with endometriosis to study the expression of them in eutopic endometrial tissue and normal uterine tissue was studied.Total of64patients with ovarian endometriosis,who had undertaken surgical treatment including laparoscopic surgery and traditional laparotomy were selected from Department of Gynecology in Tianjin Nankai Hospital.The duration time was from January2011to January2013. There are34cases in endometriosis group, selecting eutopic and ectopic endometrial tissue. In control group, there are30cases with other gynaecological disease,including oviductus tumour, oviductus coglutination. The immunohistochemistry SP method was employed to measure the expression of CyclinA and P21in eutopic and ectopic endometria.2.Total of69patients with ovarian endometriosis(Chocolate cyst),who had undertaken surgical treatment.Among them,35cases were regularly received compound Rhizome Curcumae power for3months until surgery operation(adiministration group).The rest34cases directly surgery operation without any medication(operation group).There are other30cases in control group with oviductus tumour, oviductus coglutination and so on, who had received diagnostic curettage in the endometrial proliferative phase before or during surgical operation in the same duration of hospital stay (control group). The immunohistochemistry SP method was employed to measure the expression of cyclinA and P21from ectopic and eutopic endometrium.Result:1.The immunohistochemistry SP method was employed to measure the result, through microscopy, we can observe the expression of CyclinA and P21is mainly in the uncleu of gland cell and stoma cell. Brown particulate matter is judged as positive, the proportion of expression of CyclinA is higher in eutopic endometria than in ectopic endometria in patients with endometriosis and in control group. The proportion of expression of p21is higher in ectopic endometria than in eutopic endometria in patients with endometriosis and in control group.2.The immunohistochemistry SP method was employed to measure the result, through microscopy, we can observe the expression of CyclinA is higher in eutopic and ectopic endometria in patients with endometriosis than in surgery group. Meanwhile it is higher than in control group,but not significant. The expression of P21is higher in control group than in eutopic and ectopic endometria in patients in surgery group. Meanwhile it is higher than in administration group, but not significant. And, the expression of P21is higher in eutopic and ectopic endometria in administration group than in surgery group.Coclusion:1.In control group, the expression of CyclinA is lower, but the expression of P21is higher. Meanwhile the expression of CyclinA is higher obviously in ectopic endometria than eutopic endometria in surgery group and in control group. Adversely, the expression of P21is lower in ectopic endometria than eutopic endometria in sugery and in control group. It is indicated that cyclinA and P21play roles in occurrence of the endometriosis, providing a new guide for relied studies and evidence target treatment for endometriosis.2.compound Rhizoma Curcuma powder can obviously make the level of expression of CyclinA lower, meanwhile make P21higher in both eutopic and ectopic endometrial tissue. The affection is significant in patients with mild and serious endometriosis. It is suggested that the administration of compound Rhizoma Curcuma powder to patients with in endometriosis patients before sugery is significant. It is that the poweder can decrease the lesion, which contributes to easy operation and small lesion in sugery. By administration of compound Rhizoma Curcuma powder after sugery, remant little lesion and recurrence of endometriosis can be decreased significantly. Meanwhile, chinsese medicine,which has litte bad affection, is easy to be administrated to patients in order. It plays an important role in endometriosis, especially in patients with hormone resistance.
