Objective:Establishment of a rat model for Qi stagnation and blood stasis syndrome uterine leiomyomas.Studying the reason of outcomes and the possible of mechanism for pregnancy rats,whcih is contributed by different doses of Curcumae rhizoma and Sparganii rhizoma(CRSR),by observing the changes vital signs of pregnant rats,skeleton of fetal rats,the growth and development of fetal rats after birth,and studying the Metabolic characteristics of pregnant amniotic fluid samples.Method:1.Establishment of a rat model for Qi stagnation and blood stasis syndrome uterine leiomyomas,whcih is combined with estrogen and progesterone,and multifactorial stimulation.The model rats was randomly divided into two windows,and fertilized according to 1:1 ratio of male and female.Recording the day of successful fertilization was GD0,and the next day was GD1.The first window was studyed from GD1 to GD19,and the second window was studyed from GD1 to 28 days after the birth of the pups.The both of two windows were set up with nature control group(NCG),model group(MG),postive group,high CRSR group(HCRSRG),middle CRSR group(MCRSRG)and low CRSR group(LCRSRG).The nature control group and model group was gavaged saline,the HCRSR was gavaged 66.7%concentration of CRSR,the MCRSRG was gavaged 33.3% concentration of CRSR,and the LCRSRG was gavaged 13.7% concentration of CRSR.All of thoese group the gavaged time was from GD6 to GD 15,and the giving dose was calculated at10ml/kg.2.After administration,the number of abortions,deaths and abnormal phenomena of pregnant rats in each group were observed.Pregnant rats in the first window were sacrificed at GD19,then takeing the amniotic by metabonomic analysis,examining placenta by patholog,examining half of fetuses by keletal deformities and the other half by visceral deformities.The number of stillbirths,postnatal death and growth of fetus were observed in the second window.Results:The pregnant rats came up several outcomes,which was contribute by CRSR,such as abortion and death.Besides the fetus' skeleton and visceral became deformity.The fetus birth to rats that had been gavaged medicines were grown slowly and death easily than nature control group.However the pregenant rats were not observed died and aborbtion in model group.The metabolism of amniotic fluid had changed after gavaged the medicine,contributing to Significant changes in signaling pathways,whcih had adverse effects on the environment of fetal development.It mainly involves rotein digestion and absorption,central carbon metabolism in cancer,Aminoacyl-tRNA biosynthesis,Mineral absorption,ABC transporters,Pyrimidine metabolis,Choline metabolism in cancer,Valine,leucine and isoleucine degradation and so on.Conclusion:1.The all dose of CRSR can result pregnant rats,which with Qi stagnation and blood stasis syndrome uterine leiomyomas,in poor outcomes,such as the death of pregnant rats and fetus,malformations and poor growth of the fetus.2.The blood stasis state of Qi stagnation and blood stasis syndrome uterine leiomyomas rats may be related to arginine and proline metabolism.3.The use of CRSR in pregnancy can lead to adverse pregnancy outcomes,which may be related to the disorder of protein digestion and absorption,central carbon metabolism in cancer,Aminoacyl-tRNA biosynthesis,mineral absorption,ABC transporter and so on pathway.
