Objective:Explore the relationship between early fatty liver and blood stasis syndromefrom the angle of sinusoidal endothelial cell injury and platelet activation; From the"blood stasis" angle clarifies the early law in the traditional Chinese Medicinetreatment of fatty liver, to provide the treatment and basic theory for early fatty liverwhich are more reasonable and effective.Methods:1.Preparation: The120SD rats were numbered by SPSS software randomly andprogrammed in three random numbers, also the mice were randomly divided intothree groups which are named to the normal group50, model group50andGexia Zhuyu Decoction group20. The rats of normal group are fed with normal diet,the rats of model group and Gexia Zhuyu Decoction group are fed with high fat diet.In addition, All groups of rats drank tap water.2Giving medicine in different groups: From the date of modeling, since theeighth weekend, the normal group and model group are using gastric lavaged bynormal saline, Gexia Zhuyu Decoction group are received therapeutic irrigationwith Gexia Zhuyu Decoction gastrolavage.3.Detecting indicatrix: Observed the liver function, blood lipids, liver lipids,platelet aggregation, indicators of vWF in sinusoidal endothelial cells andimprovement and therapeutical effect at the4、6、8、10、12weekends. At the4,6,8,10,12weekends, monitored the the degree of fatty degeneration by lightmicroscopy and the ultra structure pathology change of sinusoidal endothelial cellby electron microscopy dynamically.Results:1.Compared with the normal group, the rats’ weight of model group issignificantly increased. After8weeks, some rats appeared poor appetite, reduced theactivity, presented loose stools and other phenomena. 2.Compared with the normal group,4weeks later, in the model groups, rats’liver pathology showed nearly1/3of liver cells of unequal size of lipid dropletvacuoles, hepatic cord disorder, Hepatic sinus stenosis deformation by lightmicroscopy;6weeks later, Hydropic degeneration of hepatocytes, liver cells lightlystained matrix loose, lightly stained nuclei increased slightly, moderate fattydegeneration of liver cells;8weeks later, liver cells displayed severe waterydegeneration, diffused swelling induced hepatic lobules unclear, severe fattydegeneration. The cytoplasm ranging from the size of the fat vacuole. Electronmicroscope (NES). Meanwhile, since the beginning of the4th week to the end of12th week, model rats’ weight, liver weight, serum TC, TG, ALT, liver fat, plateletaggregation, vWF content increased (P<0.05).3.Compared with the model group, at the10,12weekends, platelet aggregationand vWF of Gexia Zhuyu Decoction group were significantly decreased(P<0.05).This Indicate there are certain inhibitory effect which showed that methodspromoting blood circulation and removing blood stasis on early hyperlipidemic ratplatelet hyperreactivity.4.Compared with the model group, at the10,12weekends, the fatty degeneration,liver function, elevated blood lipids and more pathologic changes of Gexia ZhuyuDecoction groups were reduced. It Indicates that the application of blood stasismethod has good effects on early intervention of fatty liver disease in rats.In Conclusion:1.After fed rats with improved High-fat diet recipes (ie normal diet of88%,10%lard,2%cholesterol) we can easily find out that fatty degeneration of the liver cellsby electron microscope in the4weekend. The liver cells showed moderate hydropicdegeneration, ranging in size within the cytoplasm of fat vacuoles, accompanied byhyperlipidemia, liver dysfunction on the6weekend and it showed severe change tothe first10and12weekend.2.High-fat diet leads that platelet aggregation rate increasing in fatty liver modelgroup, indicating the presence of high platelet reactivity in the development of fattyliver.3.Early fatty liver model group that lead by feeding high-fat diet exists Liversinusoidal endothelial cell injury, its value increased expression of vWF, electronmicroscope (NES), Indicating that the cells of hepatic sinusoidal endothelial cellinjury and specific factor vWF mediated the development process offatty liver. 4.Application of Gexia Zhuyu Decoction can improve liver function, bloodlipids, liver fat, and fatty degeneration of a model rat, also can inhibit liversinusoidal endothelial cell injury5.there is a chance that Chinese medicine blood stasis in fatty liver formationprocess, and always accompanied by the entire development process. Using bloodcirculation or blood circulation drugs for treatment may have some curative effects.
