| Background and PurposeIn China, the incidence of gastric cancer increases every year and continues ahigh level, the current treatment includes symptomatic treatment of gastric cancersurgery, radiotherapy, chemotherapy and traditional Chinese medicine, but gastriccancer often lacks clear early clinical manifestations and has a high proportion ofrecurrence or metastasis2years after surgery; gastric cancer cells are not sensitive toradiation and the tolerance is low, so chemotherapy has become the main method oftreatment. But chemotherapy has so many side effects that over the years, peoplehave been looking for new anti-cancer drugs or new combination therapy approach,hoping to reduce the toxicity of chemotherapy drugs or increase susceptibility tochemotherapy. The purpose of this study is to view Amoxicillin sodium potassiumclavulanate alone and in combination with chemotherapy drugs on gastric cancer cellsto see if it can enhance chemotherapy-induced apoptosis of gastric cancer, in order tofind a better chemotherapy drug sensitizer for patients with gastric cancerchemotherapy, and provide some theoretical basis for further exploring gastric cancercells mechanism of apoptosis.MethodGrow AGS human gastric cancer cells in vitro, detect cell morphology, draw cellgrowth curve; by MTT assay, detect AGS cells with different concentrations of thedrug in the survival and growth of the state in different time; through Transwell cellinvasion and migration experiments, test AGS cells after different concentrations of the drug in the invasion and migration of changes in the role after12h; use AnnexinV-FITC/PI double staining, detect AGS cells by flow cytometry after effects ofdifferent concentrations of the drug in the rate of apoptosis after48h.ResultAGS gastric cancer has a clear cell growth characteristics, its doubling time is20h, and its S linear growth curve trend is apparent in each period; MTT assayshowed: in the effect of different drug concentrations at different time, AGS cellgrowth inhibition effect was time-and dose-dependent manner, amoxicillin andclavulanate potassium sodium alone can cause a decline in the number of viable cells,and significant inhibited cell viability combined with chemotherapy drugs; Transwellinvasion and migration assay results showed that in different drug after12h, invasionand migration of cells are damaged, amoxicillin and clavulanate potassium sodiumalone can affect cell invasion and migration, and has a ignificant influence combinedwith chemotherapy drugs; flow cytometry results display that in the effect of differentconcentrations at48h, AGS cell apoptosis rate of amoxicillin and clavulanatepotassium increase compared with the control group, and combinated withchemotherapy drugs, its rate increase significantly than chemotherapy drugs group.ConclusionAmoxicillin and clavulanate potassium sodium alone can inhibit gastric cancercell proliferation and migration and invasion ability, and can induce apoptosis in AGScells, these effects can be enhanced when combined with chemotherapy drugs. |
