Objective To investigate the Relationship of Heat Shock Protein27(HSP27)andbiological characteristics of Prostate Cancer (PC), and explore HSP27initiallypractical value.Methods Expression of HSP27were determined by immunohistochemical method in30cases of Benign prostatic hyperplasia (BPH) and75case of PC. Analysis thecorrelation among the expression of HSP27and differentiation degree, clinical stageand serum level of prostate specific antigen (PSA) in PC. The level of HSP27weredetected by western bloting in prostatic carcinoma cell lines with different metastaticpotency. After performed siRNA to C4-2B cell line, the expression of HSP27wasdown-regulated and the Transwell test were performed to detected the changes ofinvasive and metastatic capability.Results The positive ratio of HSP27expression were65.3%(49/75)in75cases of PC,and13.3%(4/30) in30cases of BPH. And there were significantly differencebetween PC and BPH (P<0.05).75cases of PC were scored by Gleason scoresystem. The positive ratio of HSP27expression were40.0%(10/25)、68.8%(22/32)and94.4%(17/18) in75patiens of PC with Gleason2~6score、7score、8~10score,respectively. There were relationship between higher expression ratio of HSP27andhigher Gleason score in PC. And there were significantly difference among them (P <0.05). The positive ratio of HSP27expression were25.0%(3/12),59.5%(22/37),92.3%(24/26) in PC patiens with PSA<10ng/ml、10~20ng/ml、>20ng/ml,respectively. There were relationship between higher expression ratio of HSP27andhigher serum level of PSA in PC. And there were significantly difference among thethree goups with differential level of PSA in PC (P <0.05). The experiment resultsindicated that there were relationship among the expression of HSP27and TNM stages, in the clinical stage respectively for≤T2a、T2b、≥T2c among the three goupsin patiens of PC,The positive ratio of HSP27expression were14.3%(2/14)、53.3%(8/15)and84.8%(39/46). The expression level of HSP27were higher in C4-2B celllines with high metastatic ability than that of LNCaP cell lines with lowly metastaticability, and there were significantly difference between them(P <0.01). The Transwelltest showed that the invasive cell couting were higher in C4-2B-HSP27-siRNA thanthat of C4-2B and C4-2B-empty-vector, downregulation of HSP27could result intothe decreasing of in vitro invasive ability in C4-2B-HSP27-siRNA cell line(P<0.05).Conclusion1) The expression of HSP27were higher in PC than BPH, and there wererelationship between expression of HSP27and Prostate Cancer risk factors such as:Gleason score, metastasis, clinical stage and serum level of PSA in PC.2) Down regulation of HSP27by siRNA could decrease the invasive and metastaticability in PC cell lines.
