| OBJECTIVE: Studing of the factors that inference separation oftea-polyphenols from Sanye insect tea by an ion precipitationâ€solvent extract method.To optimize the extraction technology of tea-polyphenolsfrom Sanye insect tea by studing the influence factors using single factorcirculation method.To observe the tissues and ergastic substances in thepowder of Sanye insect tea with the light microscope.Method: Tea-polyphenols in Sanye insect tea were extracted by he-ating-circumfluence and test by ultraviolet and visible spectrophometry.in which it investigated influence of collectreagents and precipitants andacids conversation solution and extraction times on separation of tea-pol-yphenols respectively with the single factor method.The microscope wasused to observe the tissues and ergastic substances in the powder of Sany-e insect tea after mounting with water and after mounting with chloral hy-drate by heating respectively.Result: Comet assay datas demonstrated that the best extracting con- dition was with80%acetone as collectreagents and optimum separationwith ZnCl2as precipitant and hydrochloric acid as conversion solution.The extraction rate of polyphenols from Sanye insect tea by better techn-ology was14.57%.There were non-glandular hairs and brown blocks andsolitary crystals and vessels and starch grains in the powder of Sanye ins-ect tea.Conclusion: In this study, the extraction technique of polyphenolsfrom Sanye insect tea was studied by an ion precipitation-solvent extractmethod,The optimum parameters were obtained by the single factor test.The optimum parameters were as follows:80%acetone as the extractsolvent,ZnCl2as the precipitant,2mol/L HCl as the dissolving solutionand once of the extract times of the acid aqueous solution by Ethylacetate reaching95%.Under the optimized extract procedure, theextracting rate of tea polyphenols to Sanye insect tea was up to14.57%(g/g)with the RSD (n=3) of0.4%. The results demonstrated that thepresent extract procedure of tea polyphenols from Sanye insect tea wasstable with the high extracting rate and the desired repeatability.Inaddition,the identification of powdered Sanye insect tea showed that itcontains solitary crystal, starch grains contents, et al. |
