Objective:To investigate the polymorphisms of tumour necrosis factor-?(TNF-?)gene promoter region at position rs1799724(-857,C/T)?rs1800630(-860,C/A)and rs1799964(-1031,T/C)in xinjiang uygur with vitiligo;and to explore the association between TNF-? gene promoter region at position rs1799724 ? rs1800630 and rs1799964 loci polymorphisms in xinjiang uygur with the pathogensis of vitiligo.Methods: Study of the xinjiang uygur divided into vitiligo group of 316 cases(140males,176 females)and with healthy control group of 330 cases(199males,131females).Geno-mic DNA were extracted from peripheral whole blood cell and gene sequencing methods.To determined of the TNF-? gene promoter region at position rs1799724 ? rs1800630 and rs1799964 loci polymorphisms.Apply SPSS17.0 statistical analsis software to read gene alleles and genotypes.To compared that each group of the frequency of genotype and allele which between vitiligo group and healthy control group,vitiligo clinical phenotypes with chi-square test.Results:1.In xinjiang uygur vitiligo group and healthy control group,TNF-? gene promoter region at position rs1799964 loci polymorphism exists the three genotypes TT(61.07%,60.61%),TC(35.12%,33.33%),CC(3.80%,6.60%)and the two alleles T(78.64%,77.27%),C(21.36%,22.72%)in xinjiang uygur population;TNF-? gene promoter region at position rs1800630 loci polymorphisms exists the three genotypes CC(68.67%,72.12%),CA(30.38%,26.06%),AA(0.95%,1.81%)and the two alleles C(83.86%,85.15%),A(16.14%,14.85%)in xinjiang uygur population;TNF-? gene promoter region at position rs1799724 exists the three genotypes CC(72.15%,70.0%),CT(25.63%,26.97%),TT(1.10%,3.03%)and the two alleles C(84.97%,83.48%),T(15.03%,16.52%)in xinjiang uygur population.2.There were no statistical significance in the allele and genotype frequencies of TNF-? gene promter at position rs1799724?rs1800630 and rs1799964 between xinjiang uygur vitiligo group and healthy control group(P?0.05).3.In xinjiang uygur vitiligo group,there were no statistical significance in the genotype and allele frequencies of TNF-? gene promoter region at position rs1799964 and rs1800630 with the clinical phenotypes in vitiligo group(P>0.05).But the frequency of CT+CC genotype with rs1799724 locus in focal vitiligo group was significantly higher than other types group(?2=22.889,P<0.05);the T allele frequency of rs1799724 locus in focal vitiligo group was significantly higher than other types group(?2=22.651,P<0.05).The frequency of CT+CC genotype with rs1799724 locus in active vitiligo group or family history vitiligo group was significantly higher than stable vitiligo group or no family history group(?2=36.114,P<0.05,OR =5.035,95% CI: 2.899~8.743;?2 =15.527,P<0.05,OR =5.644,95% CI: 2.189~14.651);the T allele frequency of rs1799724 locus in active vitiligo group or family history vitiligo group was significantly higher than stable vitiligo group or no family history group(?2=35.109,P<0.05,OR =3.691,95%CI: 2.353~5.790;?2 =2.390,P<0.05,OR =12.995,95% CI: 1.822~9.999).Conclusion:?There are TNF-? gene promoter region rs1799724,rs1800630 and rs1799964 loci polymorphisms in xinjiang uygur population,and there were no statistical significance between the two groups.?TNF-? gene promoter region rs1799724 loci polymorphism allele T may be genetically susceptible to the progression of vitiligo,which in active,focal,family history with xinjiang uygur.?There are no association between TNF-? gene promoter region rs1800630 and rs1799964 loci polymorphisms and vitiligo clinical phenotypes in xinjiang uygur.
