ObjectiveTo study the expression and clinical significance of glial fibrillary acidic protein (GFAP) and CD57in pleomorphic adenoma (PA) and adenoid cystic carcinoma (ACC).For further evaluating the role of glial fibrillary acidic protein (GFAP) and CD57in differentiating PA from ACC.MethodsSubjects selected from the archived paraffin-embedded salivary gland tumors which were surgical excision by oral and maxillofacial surgery of First Hospital of Shanxi Medical University by the year2006-2009:(1)8cases of salivary gland normal tissue,(2)20cases of PA,(3)16cases of ACC. Immunohistochemistry method was used to measure the expression and localization of GFAP and CD57in tissues of salivary gland, calculation of values GFAP and CD57score results, the data was analyzed by SPSS(17.0) software.Results1.In our study, all8cases with cytologic diagnoses of salivary gland normal tissue were negative for GFAP, and3of8cases were positive for CD57; All20cases with cytologic diagnoses of PA were positive for GFAP, and18of20cases were positive for CD57; All16cases with cytologic diagnosis of ACC were negative for both GFAP. And13of16cases were negative for CD57. The GFAP positive rate in PA was100%. There is no GFAP expression in ACC and normal parotid gland tissue. By Kruskal-Wallis test, In the three groups GFAP expressions in the normal parotid gland, PA and ACC were Statistically significant (x2=9.964, P=0.007);2.CD57expression in the normal parotid gland tissue was37.5%, in PA was90%, in the ACC, the positive expression rate was18.7%.By Kruskal-Wallis test, CD57expression in the three groups were Statistically significant(x2=6.248, P=0.044);3. by Spearman’s correlation analysis, the P <0.05, so that GFAP and CD57expression in the three groups are related.ConclusionsIn sum, Our results show that.1.The high GFAP positive expression rate in the PA, and in the adenoid cystic carcinoma is almost no expression, the reason may be pleomorphic adenoma of the formation of the reason may be the formation of immature cartilages in PA;2. The high CD57positive expression rate that was90%in the PA, and in the adenoid cystic carcinoma was just18.7%.,which can be speculated that CD57is effective in tell benign from malignant parotid gland tumors, and it was a important tole in judging prognosis of the tumor.3. A large number of analysis shows:positive staining for GFAP and CD57serves as a useful adjunct for the diagnosis of PA and helps to reduce the uncertainty in challenging cases.
