ObjectiveColorectal cancer is one of the common gastrointestinal cancer, its morbidity andmortality has been increased gradually in recent years, and therefore its preventionhas become the focus of research and clinical work. P-21activated kinase4(PAK4) isa newly discovered multifunctional effector which can regulate a variety of cellularactivities including cell migration, cytoskeletal movement, cell proliferation,apoptosis, angiogenesis and genome transcriptional regulation, etc, when activated byits upstream signaling pathways. It plays a crucial roal in the normal cellular activities,once PAK4has abnormal expression which will cause tumorigenesis and tumorprogression. Recently,PAK4has been shown to overexpress in a variety of tumorsand tumor lines, such as breast cancer, gastric cancer, prostate cancer, ovarian cancer,and it is also closely related to the occurrence and progression of these tumors, but thecorrelation studies in colorectal cancer is fewer. This paper aims to investigate theexpression of PAK4gene in colorectal cancer and its relationship withclinicopathological features through experiment which in order to evaluate thepossibility of being a new tumor marker or prognostic factor in colorectal cancer.MethodsThe mRNA expression of PAK4gene was detected by Real-time quantitativereverse transcription-polymerase chain reaction (Real-time RT-PCR) in65cases of colorectal cancer tissues and paracancerous normal tissues and apply statisticalmethods to analyze its relationship with clinicopathological features. Statisticalmethods use independent samples T-test and one-way ANOVA, P<0.05indicatessignificant difference.ResultsReal-time RT-PCR outcome analysis revealed that the expression of PAK4gene in46cases of colorectal cancer tissues compared with paracancerous normaltissues(More than5cm from cancer tissue, the pathological examination withoutcancer invasion) were greater than1,19cases were less than or equal to1, the totalamount of relative expression was2.203±2.571, the difference was statisticallysignificant (P <0.05); there was no significant correlations between the expression ofPAK4and sex, age, tumor location, tumor size, pathologic differentiation degree (P>0.05), but it’s closely related to the depth of tumor invasion, lymph node metastasis,TNM stage (P <0.05).Conclusions1. PAK4gene is overexpressed in colorectal cancer tissues.2. Upregulation of PAK4gene in the colorectal carcinoma suggests that PAK4may be involved in the carcinogenisis and development of colorectal cancer, and it isexpected to become a new tumor marker and prognostic factor for colorectalcarcinoma.
