| Objective To investigate the expression of Abl—interacting protein1(ABI1) in normal gastric mucosal cell line GES-1and gastric cancer cell line SGC-7901, and the mechanism for growth and progress of gastric cancer.Methods Observe the localization of ABI1in normal and cancer cell lines by immunohistochemistry and immunofluorescence. Compare the gene expression of the two types of cell lines at the protein and mRNA level by Western blot and real-time q-PCR, analyzing the expression pattern of candidate cell lines.Results①Immunohistochemistry and immunofluorescence showed that ABI1in normal gastric mucosa cell line GES-1was mainly expressed in the cytoplasm and in gastric cancer cell line SGC-7901was mainly expressed in the nucleus, the luminous intensity of GES-1cell lines is higher than that of SGC-7901cell lines. The positive staning area of ABIl in cell line GES-1and cell skeleton are basically the same.②Western blot and Real time quantitative PCR showed that ABI1protein expressed both in GES-1and SGC-7901cell lines. The expression levels of ABI1protein and mRNA of cell line SGC-7901was lower than cell line GES-1(P<0.01).Conclusion ABI1may be involved in the occurrence and development of gastric cancer by down-regulating protein expression, may be a new tumor marker. |
