Objective: Research the affect of Selaginella and the estrogen-like effective part ofrehmannia glutinosa libosch on osteoporosis in ovariectomized rats. and the preliminarystudy of its osteoporosis intervention mechanism.Methods: Wistar female rats were carried out the castration to remove both ovaries (exceptthe sham group) to establish the rat model of postmenopausal osteoporosis. The Selaginellaresidue water extraction after refluxing extracted by95%ethanol (SEW), the freshrehmannia glutinosa libosch alcohol extraction refluxing extracted by95%ethanol (FREE),the decoction of fresh rehmannia glutinosa libosch by aqueous extracted (FREW), theprocessed rehmannia glutinosa libosch alcohol extraction refluxing extracted by95%ethanol(DREE), the decoction of dried rehmannia glutinosa libosch by aqueous extracted beadministered after surgery to therapeutic intervention, continuously administered40weeks.Ts were sacrificed after administered40weeks and weighed the rats and organs, calculatherated the cardiac index, liver index, spleen index, lung index, index of kidney, adrenal index,thymus index and uterus index by the ELISA detection kit; Measured the rats right femurbone density by dual energy X-ray absorptiometry, Rat Whole Body V5.73critters analysissoftware; measured the rats right femur bone biomechanical properties (maximum load,maximum displacement, elastic modulus, stiffness, maximum energy absorption) by thethree-point bending test; analyzed the level of alkaline phosphatase (ALP), calcium (Ca),phosphorus (P), blood glucose (BG), total cholesterol (TC), triglyceride (TG), high densitylipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) in the serumof rats by Automatic biochemical analyzer; detected the level of estradiol (E2), parathyroidhormone (PTH), tartrate-resistant acid phosphatase (TRAP), osteocalcin (BGP), calcitonin(CT) and I procollagen carboxyl terminal peptide (PICP) in the serum by ELISA detectionkit; observed the hippocampus, pituitary, hypothalamus, uterus, skeletal muscle, right femur,liver, heart, liver, spleen, lungs, kidneys, adrenals, pancreas, thymus, thyroid, stomach, smallintestine, abdominal aortic pathological by light microscopy. During of the20weeks afteradministration, gave the rats tail vein blood, analyzed the alkaline phosphatase (ALP), bloodglucose (BG), total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol(HDL-C), low-density lipoprotein lipoprotein cholesterol (LDL-C) levels in the serum byAutomatic biochemical analyzer; detected the serum estradiol (E2). Results: Compared with the model group, the extraction of Selaginella&rehmanniaglutinosa libosch can increase the level of castration in vivo E2and CT (P <0.05) in rats;deduce serum BG、TC、TG、HDL-C、LDL-C、ALP、TRAP、BGP、PTH、PICP level (P<0.05); significantly increased stiffness (P<0.05). HE staining showed that the extraction ofSelaginella&rehmannia glutinosa libosch can inhibit endometrial atrophy and loss, able toimprove the relative expansion of rat bone marrow cavity and trabecular thinning, sparse; noeffect on other organs.Conclusion: the extraction of Selaginella&rehmannia glutinosa libosch can regulateglucose and lipid metabolism in rats with osteoporosis after menopause, can increaseestrogen levels in the serum of ovariectomized rats, improve bone biomechanical properties,and have a certain regulation on the biochemical markers of bone metabolism, improvepathological uterus and femur in ovariectomized rats, suggesting that the extraction ofSelaginella&rehmannia glutinosa libosch have a certain intervention on osteoporosis inovariectomized rats.
