Rehmannia rehmanniae was first recorded in the book of Shen Nong's herbal classic,and is listed as the top grade.It is one of the famous "four huaiyao" in China.It has been used to treat diseases fo r thousands of years,and has been cultivated in many provinces and c ities in China.Genuine herbs are mainly produced in Wuzhi,Xiuwu and Wenxian of Henan Province.Rehmannia glutinosa is a fresh or dried root tuber or processed product of Rehmannia glutinosa libosch.The order of medication was fresh Rehmannia glutinosa,raw Rehmannia glutinosa and cooked Rehmannia glutinosa.There are obvious differences in the efficacy among the three: fresh Rehmannia glutinosa can clear heat and produce saliva,cooling blood and hemostasis.Raw Rehmannia glutinosa can clear heat and cooling blood,nourishing yin and produce saliva.Cooked Rehmannia glutinosa nourishes Yin and blood,enriches essence and fills marrow.Rehmannia glutinosa is involved in one fifth of the prescriptions in the Pharmacopoeia of the People's Republic of China published in the 2015 edition.As a traditional medicine in China,traditional Chinese medicine is mainly taken orally,which enters into the body through the mouth,enters the blood circulation system after being metabolized by the digestive system,interacts with the target of the body at a specific location,and then excretes the drug through the metabolism of tissues and organs in the body.However,because traditional Chinese medicines mainly come from animals and plants,most of them have biological diversity,variability,complexity of components and other characteristics.When drugs enter the body,experienced the process of absorption,metabolism,distribution,excretion and interac tion between components,the composition of the body becomes more complex and diverse.In addition,due to the toxic and side effects of drugs,the concentration of drugs and the amount of metabolit es in the body are generally very low.These factors increase the difficulty of traditional Chinese medicine research and also become the main obstacles to the internationalization of traditional Chinese medicine.With the development of modern science and technology,analytical detection technology has been constantly improved.The liquid chromatography-tandem mass spectrometry(lc-ms)has the characteristics of high sensitivity,high accuracy and high selectivity,which make the analysis and detection of biological samples possible.Therefore,raw and cooked Rehmannia glutinosa were studied in this article.Firstly,the extract ion process was investigated.Secondly,the chemical constituents and metabolic processes in vivo were studied by liquid chromatography-mass spectrometry(LC-MS)to provide the reference data of pharmacodynamics for their further study.Study on extraction technology of raw and cooked Rehmannia glutinosa.In this study,the extraction technology of raw and cooked Rehmannia glutinosa was optimized with the mulberry glycoside extraction rate as an index.The extraction process was mainly optimized by ultrasonic-assisted ethanol reflux method,and combined with high performance liquid chromatography determining the content of mulberry glycoside.By carrying out the single factor experiment,we investigated the effects of factors of ethanol concentration,liquid-material ratio,extraction time,extraction temperature,ultrasound power on the extraction rate of mulberry glycoside in raw and cooked Rehmannia glutinosa.On this basis,the extraction process was optimized by using response surface methodolog,and bas ed on the experimental results,the changes of the content of mulberry glycoside before and after processing of Rehmannia glutinosa were compared to provide reference for the control of processing parameters and the guarantee of medication efficacy in the processing of Rehmannia glutinosa.Twenty compounds were identified from the ethanol extract of raw Rehmannia glutinosa,including 10 iridoid glycosides,5 phenylethanoids and 2 nucleosides,and 17 compounds were identified from the ethanol extract of cooked Rehmannia glutinosa,including 7 iridoid glycosides,5 phenylethanoidosides,3 nucleosides and 2 organic acids.Study on the serum drug composition of raw and cooked Rehmannia glutinosa.Rats were used as experimental subjects,and the alcohol extracts of raw and cooked Rehmannia glutinosa were used as drugs in this study.After oral administration by gavage.Detecting by collecting the rats' plasma samples,gradient elution with acetonitrile-0.1% formic acid water as mobile phase in ACQUITY UPLC BEH-C18 column(2.1mm x 50 mm,1.7?m)and under the mode of electrospray linear ion trap mass spectrometry.Eight prototype compounds were identified from rat plasma.Conclusion: The optimum extraction technology of raw and cooked Rehmannia glutinosa was elaborated in this paper.The chemical constituents of the alcohol extracts of raw and cooked Rehmannia glutinosa were explored.The changes of the alcohol extracts of rraw and cooked Rehmannia glutinosa after oral administration were described.It provides useful data for the study of extraction technology of raw and cooked Rehmannia glutinosa.At the same time,it provides a good chemical basis for raw and cooked Rehmannia glutinosa in the aspects of clinical safety evaluation,pharmacological and pharmacodynamic research,quality control and so on.
