The traditional chineese medicine has been used by chinese for thousands of years. Manyclassic prescriptions have been studied that they have an anti-tumor efficacy. The clinicalmedicine found that Yiyuanyiliu Decoction has an effect of anti-tumor in vivo, so this articlestudies its anti-tumor effect and expores its active constituents.The continuous lavage for16d, we found that the anti-tumor rate of YiyuanyiliuDecoction was22.15%. Yiyuanyiliu Decoction can increase body weight in tumor-burdenedmice, improve appetite, perfect life quality and extend the mice survival. HE staining showeddrug group was obviously a nuclear pyknosis and patchy necrosis. Immunohistochemicaldisplayed that the MVD of medicine group significantly reduced. Immune index displayedthat the spleen index, thymus index and white blood cells were significantly activated. Thereare significant increasement in contents of IL-2, TNF alpha, IgA, IgM and IgG. Meanwhile,the fuctions of macrophage and lymphocyte were also enhanced. In conclusion, YiyuanyiliuDecoction has an effect of anti-tumor in vivo.Second, we got1~5extract components from Yiyuanyiliu Decoction by organic solventextraction. We also got6~14extract components from Yiyuanyiliu Decoction according todifferent target products. We wanted to find material foundation of immunoregulation inYiyuanyiliu Decoction. In order to screen effective constituents, T and B lymphocytes wereused as screening model. We finally got ideal group, named component6. We found thatcomponent6can not increase the numbers of white blood cells and macrophage phagocytosis.The experiments of IL-2and TNF alpha showed that the immunoregulation effect ofcomponent6can locate in them. The experiment on the exchange in contents of T and Blymphocytes showed that the immunoregulation of component6can also locate inlymphocytes.Finally, we acted A549cell, Bel7402cell, EA.hy926cell and HMEC cell as screeningmodels to screen anti-tumor components. We got ideal component, named component7,through the LDH experiment. We explored mechanisms of component7on A549cell andEA.hy926cell. We found that component7under40μg/mL can not result in cell death withtrypan blue staining method. Flow cytometry instrument detection showed component7couldchange the cell cycle and cause the cell apoptosis. Hoechst33342also showed thatcomponent7can cause cell apoptosis. We found that as the higher of componentconcentration, caspase3had higher activaty, and the ratio of Bax and Bcl-2was also raised. Itis an illustration that component7has the function of apoptosis.Frist, we verified the anti-tumor effect of Yiyuanyiliu Decoction. Then we selected theimmune component and analyzed its efficacy. We selected anti-tumor components by thebiological activity and toxicity of extract. Finally, we discussed mechanisms of component7.This topic provides experimental data and theoretical basis for clinical efficacy of YiyuanyiliuDecoction.
