Objective:To explore the effects fluctuant high1blood glucoseand Calcitonin gene-related petide on kidney in diabetic rats.Methods:The male Sprague-Dawley(SD)rats randomly weredivided into4groups:control group,control fluctuant bloodglucose,stable high blood glucose group and fluctuant highblood glucose group.Diabetes rats were induced byintraperitoneal injection of STZ.and the fluctuant high bloodglucose animal model was induced by subcutaneous injectioninsulin and intragastric administration glucose at differenttime points every day.9weeks later weight、kidney index(KI)、mean blood glucose(MBG)、standard deviation of blood glucose (SDBG)、largest amplitude of glycemic excursions(LAGE)、24h urinary albumin、 blood urea nitrogen (BUN)、 serumcreatinine(Cr)、HbA1c、blood Calcitonin gene-related peptide(CGRP) were determined, Observing change of kidneymicrostructure by light microscope.Results:Compared with group A and group B, group C and groupD have higher KI、MBG、SDBG、LAGE、24-hour urine microalbumin、BUN、Cr, and HbA1c and lower body mass and CGRP (P<0.05).Further analysis of group C and group D shows that group D hashigher KI、SDBG、LAGE、24-hour urine microalbumin、BUN, and Crand lower body mass、 MBG and CGRP (P<0.05). Pearsoncorrelation analysis demonstrates that SDBG and24-hour urinemicroalbumin,BUN,Cr have positive correlation.Correspondingly, CGRP and24-hour urine microalbumin, BUN,Crhave negative correlation.Conclusion:Fluctuant high blood glucose and the decrease ofthe levels of CGRP were associated with the development ofdiabetic nephropathy(DN).
