The Anti-Tumor Efficacy Study Of Andrographis Effective Parts(AEP), Tgf-β1Regulator

Andrographis paniculata(Burm.f.)Nees was found in India and planted in east, middle south and southwest China. It is used extensively as an antipyretic, anti-inflammtory and antivirus medicine for a long time. It is known as "Chinese medicine antibiotics". In recent years, many studies have shown that, in the represented by paniculta lactone diterpene lactone class composition has a broad spectrum of anti-tumor activity. Andrographis effective parts (AEP) is the supramolecular technology to enrichment of andrographis alcohol extract, andrographis of supramolecular extracts, including andrographolide (A), dehydroandrographolide (DDA),14-deoxyandrographolide (DA).This topic is to research effective of AEP on4T-1breast cancer cells in mice, LLC lung cancer in mice and A549lung cancer cell in the body. Study the inhibitory effect on the immune adjustment factor TGF-β1, to explore the antitumor effect of AEP.In vitro studies, AEP as model drug, with breast cancer cells4T-1in mice and lung cancer cells LLC in mice as tumor models, using tetramethyl azo thiazole blue (determined by MTT method), inspects the drug to the tumor cell proliferation inhibition.Results showed that AEP for the growth of tumor cell proliferation has obvious inhibitory effect, and have time and dose dependence, there are significant difference compared with the blank control group (P<0.05), and the IC50of4T-1cell is (47.33±1.36)μM (24h) and (26±0.93)μM (48h), the IC50of LLC cells is (39.6±1.27)μM (24h) and (31.7±1.01)μM (48h), the antitumor function of AEP on the two kinds of cells were stronger than A.In vivo study, AEP as drug model, with breast cancer cells4T-1inoculated BABL/C mice and lung cancer cells LLC inoculated C57BL/C mice as tumor models, with the tumor inhibition rate of life extension, viscera index, serum biochemical index (SOD and LDH, y-GGT, AKP,5-NT), pathological section as indexs, through gastric drug investigation of AEP for4T-1tumor and LLC tumor inhibition.Results show that the AEP for solid tumor4T-1and LLC has good inhibition, the tumor inhibition rate of4T-1reached68.82%, LLC,74.34%;With physiological saline group than mice significantly prolong the survival period, improving the mental state of mouse;The viscera index of AEP group compares to the normal control group having no significant difference. It shows that AEP have no obvious damge effect to the organ organization;Serum biochemical index SOD activity in the drug group compared with normal saline group have significantly improved, drugs can improve the body’s antioxidant system, serum biochemical indicators of the four specific tumor results compared with negative control group also have varying degrees of lower, prompt drugs on the body in the later stages of the development of tumor metastasis and toxins to damage organs have the effect of the treatment and improve;Pathological analysis showed that tumor cells in tumor tissue and metastasis viscera tissue morphological changes, cell pycnosis, cracking necrosis and inflammation.In vivo experiments, AEP as model drug, BABL/C nude mice inoculated anthropogenic lung cancer cells A549tumor model, through lavage, inspects the drug to the tumor inhibition.Results show that the AEP has good inhibition to the A549, the tumor inhibition rate of A549reaches78.75%, and the mental state to medicine group mice compared with saline group improved significantly.Growth factors in the mice in the study of TGF-β1, AEP as drug model, breast cancer cells4T-1in mice and lung cancer cells LLC in mice for tumor model, by transforming growth factor determined by MTT test and mice TGF-β1kit, drugs on tumor cells in the process of growth and development of TGF-β1secretion.Results show that the AEP can effectively inhibit tumor cell secretion of TGF-β1, have very good immune regulating effect.This topic found that the effect of AEP on breast cancer cells4T-1in mice and lung cancer cells LLC in mice has good inhibition both in vitro and in vivo, the anthropogenic A549lung cancer cell also has good inhibition, the tumor cell growth factors secreted TGF-β1also has significant inhibitory effect on the body has very good immune adjustment.
